Lagerstroemia is Liliaceae,Lagerstroemia genus.It is a deciduous shrub or small tree.Lagerstroemia originated in China,one of China's important traditional flower,with 1500 years of cultivation history.Compared with other ornamental plants,Lagerstroemia tree is beautiful,flowers in the summer flowers,long flowering,colorful,anti-pollution ability,in the garden has a very wide range of uses,with important garden applications and ornamental value.The varieties selected for this test is the American Black Diamond Lagerstroemia,the characteristics of this species is the leaves appear purple of the four seasons,almost close to black.Other varieties of Lagerstroemia Although the flowers have a strong ornamental,but in the view of foliage in this area is still inadequate.Black Diamond Lagerstroemia as a new generation of flowers,foliage in one of the new varieties,just to fill the vacancy.Making the leaf color to become the following dry skin,color,plant type after another very important ornamental traits.However,because the species of Lagerstroemia is relatively rare,using the traditional breeding methods such as sowing,it will make it difficult to maintain excellent traits.Can maintain good traits of cutting propagation method is too slow,it is difficult in the short term to achieve a large number of breeding to meet the needs of the market.Therefore,the object of this study is American Black Diamond Lagerstroemia,its rapid propagation system for the establishment,not only have some academic value,but also to meet the black drilling Lagerstroemia rapid factory production,with the practical needs of the market to meet the needs.In this experiment,the stem of Lagerstroemia indica was used as the research material.From the selection of basic culture medium,the most suitable sterilization method in the first generation culture,the optimum amount of hormone used in the first generation,the most suitable culture medium and hormone Dosage,as well as the most suitable rooting culture medium and culture hormone types and dosage selection of several aspects of the study,and ultimately to screen out the most excellent combination of technology.Explosive test of the explants showed that the first 70%alcohol disinfection for 30s,and then 10%NaClO solution disinfection 10min more suitable for this test selected materials.At this time the pollution rate was 14.43%,browning rate of 23.33%,and other ways of pollution rate and browning rate significantly different.PPM,as a broad spectrum antimicrobial agent,can eliminate endogenous contamination of explants at higher concentrations.The addition of PPM can significantly reduce the pollution rate in the first generation culture process,which is beneficial to induce the axillary buds of Lagerstroemia indica.In the future tissue culture test,can be appropriate to use PPM,the smooth progress of the test have some help.The results showed:for the sprouting of stem segment in vitro,the best culture medium formula was MS+6-BA1.5mg/L+NAA0.5mg/L+PPM2.0mg/L,with differentiation rate of 76.33%.The subcultured test showed that the best subculture medium was MS +2.0mg/L 6-BA,and the growth peak of the crape myrtle was the highest and the highest was 7.26.This type of cluster buds is most conducive to rapid propagation.The results showed that the regenerated seedlings of Lagerstroemia indica could induce rooting in the rooting medium supplemented with different concentrations of IBA and NAA,but the rooting and rooting rate were significantly different.In the whole rooting orthogonal test,1/2MS +0.5mg/LIBA medium induced the best rooting effect,the highest rooting rate,100%,thick roots,growth is also good.The survival rate of domesticated and transplanted tissue culture plants can reach 72%and above.
