| Lonicera japonica Thunb.is a perennial semi-evergreen green woody vine plant.Honeysuckle is the dry flower buds or early flowers of Lonicera japonica Thunb..Honeysuckle has the functions of heat-clearing and detoxifying,cooling and cooling,anti-virus,etc.It has a wide range of uses in food,health care products and daily chemical products.At present,there are long-term asexual reproduction,serious pests and diseases in the production of honeysuckle,which cause the variety to degenerate,yield and quality.It is necessary to solve these problems by creating new varieties and breeding new varieties.In this experiment,the specificity of new varieties of Lonicera japonica L.and the creation of new germplasm were studied by means of scanning electron microscopy,optical microscopy,ultraviolet spectrophotometer,enzyme-linked immunosorbent assay(ELISA),transcriptome sequencing,systematic breeding and cross breeding.The mechanism of the growth of the epidermis of the honeysuckle flower buds,the color change,and the delayed flowering specificity of the flower buds were discussed.2 new honeysuckle lines were selected and 3 new germplasms were created.The research results are to further study the molecular mechanism of the specificity.The breeding of new varieties has laid a solid theoretical,technical and material basis and has important academic and applied value.1.The results of the study on the flower buds or flower epidermis of 4 varieties of Lonicera japonica Thunb.showed that there were significant differences in the density and length of the epidermis in different periods and different varieties.The density and length of the epidermis of Yujin 1 were the highest.The glandular hairs are multicellular glandular hairs,and the heads contain pigments.The non-glandular hairs are single-cell thick-walled non-glandular hairs.There were 3 genes expression levels of Damaohua and Yujin 1,which were consistent with the change in the number and length of epidermal hairs,and 5genes of them had opposite changes.2.The results of the study on the distribution,content and color of pigments in Damaohua and Yujin 2 corollas showed that the color of the corolla in the two varieties of Lonicera japonica Thunb.,the color of the corolla in the same period and the color of the cross section of the corolla in different periods were different.The content of chlorophyll,carotenoids and flavonoids changed significantly.The distribution of pigment and its content were the key factors affecting Lonicera japonica Thunb.color.3.The results of studies on the duration and endogenous hormones(IAA,ZR,GA3,ABA)of Damaohua and Telei 1 showed that the main factors affecting the length of the Telei 1 white period were high level IAA and low level ABA.There are also high ratios of IAA/ABA,ZR/ABA,IAA/GA3 and low ratio ABA/GA3,ZR/IAA,which provide theoretical support for the use of exogenous hormones in the regulation of Lonicera japonica Thunb.flowering and prolonging the harvesting period.4.The results of the research on the breeding system and pollination characteristics of Lonicera japonica Thunb var.chinensis(Wats.)Bak showed that its breeding system is a mixed mating system,which mainly carries out cross-pollination by wind and insects.It belongs to gamete type self-incompatibility,and the self-sufficiency rate is low.5.In this experiment,2 new lines were obtained through systematic breeding,and three excellent new germplasms and 1,303 seeds of four hybrid combinations were obtained by cross breeding,which laid a material foundation for identification of new varieties,selection of new lines and creation of new germplasm. |
