Pueraria lobata(Willd.)Ohwi is a leguminous Pueraria plant with a wide range of uses and economic value.With the increase of market demand in recent years,the selection and breeding of excellent varieties is the primary task at present.In this experiment,ISSR molecular markers combined with phenotypic traits of Kudzu were used to analyze the differences between individuals and screen excellent plants with high quality and high yield,which provided some references for molecular marker-assisted selection breeding to improve the yield of Kudzu and related agronomic traits.In this experiment,137 Kudzu were analyzed and studied by phenotypic traits and ISSR molecular markers respectively to provide theoretical basis for germplasms of kudzu and cultivation of new varieties.The results are as follows:1.The 25 phenotypic traits were selected for field investigation and determination of137 Kudzu of the experiment.The results showed that there were significant differences in 16 morphological traits.The variation range of coefficient of variation was17.55%-174.04%,and the average coefficient of variation was 71.31%.The variation sources of morphological characters of 137 Kudzu were mainly root-shoot ratio,fresh weight of above-ground,single root weight and so on.Among the five physiological indexes,the variation of transpiration rate and relative chlorophyll content is obvious,with the variation coefficients of 59.41% and 49.53%,respectively,and the average variation coefficient of 36.00%.The variation mean value of six nutrient composition indexes of Kudzu is 19.97%,and the variation range is 9.42%-32.93%,of which the variation degree of P content and Ca content is large,and the variation coefficient is32.93% and 28.16% distinctly.2.ISSR-PCR amplification was carried out on 137 Kudzu by 22 primers,and the amplified products were detected by agarose gel electrophoresis.A total of 62 bands were amplified by 22 primers,of which 58 bands were polymorphic,the percentage of polymorphism was 92.05%,with an average of 2.64 bands per primer.Among them,primer UBC826 amplified the most bands,with 5 amplified bands,polymorphic bands5,polymorphism is 100%.Primers UBC829 and UBC853 have the least amplified bands,each with one polymorphic band and 100% polymorphism.3.Through PCR amplification of 22 primers,the phenotypic traits of 137 Kudzu in different periods were analyzed by SPSS17.0 software according to the amplified products by variance and multiple or T-test analysis.The results showed that there were significant differences or extremely significant differences in the traits of 137 Kudzu at different growth stages.The main differences were root-shoot ratio,fresh weight of aboveground parts,stem diameter,internode length,etc.According to the amplification results,primers with different characters are USB810,USB827,USB829,USB836,USB840,USB846 and USB851.Stem diameter(SD),leaf width(LW),branch number(SN)and above-ground fresh weight(GFW)can be used as the basis for selection of excellent plants.Primer USB829 is associated with stem diameter(SD),USB840 is associated with internode length(IL)and net photosynthetic rate(Pn),and primer USB846 is associated with leaf width(LW),relative chlorophyll content(SPAD),root diameter(RD),Ash(ASH)and phosphorus content(P).
