Pueraria lobata(wild)Ohwi 'Dabashan' was main varieties of kudzu vine industries in Sichuan province, it has broad habitat, high output and rapid growth, rich in starch and tolerance to drought and barren. Colchicine and two kinds of herbicide may change the morphological characteristics of plant, and then impact the physical activities to improve the yield and quality. In order to further increase the production and quality, expand the potential usably of kudzu vine industries, this study used Colchicine, Trifluralin, Pendimethalin dispose the multiple shoot clumps of kudzu in vitro with two methods, soaking and mixed culture. Study the impact of height, blade area, density and area of stomas and the number of chromosome in root tip cells after 40 d, proliferation of two generations dealt with three chemical agents and the rooting of tube seedlings. The results showed that:(1) After 40 d of treatment with Colchicine, the fatality rate was 5.24% higher than the control group, the height was 41% reduced, the blade area was increased by 44%, the density of stomas was reduced by 55% and the area of stomas was increased by 48% when treated 24 h with 0.2% in soaking. In mixed culture,0.01%?0.03%?0.05% were all not the lethal dose to plant, the height was 50% reduced, the blade area was increased by 54%, the density of stomas was reduced by 30% and the area of stomas was increased by 37% at the concentrate of 0.05%.(2) After 40 d of treatment with Trifluralin, the fatality rate was the lowest which reached 7.04% when treated 12 h with 0.01% in soaking, the time of treatment had no significant impact on height, density and area of stomas. With the concentrate of 0.01% in soaking, the height was 3% reduced, the blade area was increased by 50.7%, the density of stomas was reduced by 35% and the area of stomas was increased by 38.9% when compared to control group. In mixed culture,0.001%?0.003%?0.05% were all not the lethal dose to plant, the height was 22.06% reduced, the blade area was increased by 36%, the density of stomas was reduced by 16.4% and the area of stomas was increased by 21.3% at the concentrate of 0.005%.(3) After 40 d of treatment with Pendimethalin, the fatality rate was the lowest which reached 16.67% when treated 12 h with 0.01% in soaking, the time of treatment had no significant impact on height, density and area of stomas. With the concentrate of 0.01% in soaking, the height was 9.9% reduced, the blade area was increased by 30.5%, the density of stomas was reduced by 16.4% and the area of stomas was increased by 37% when compared to control group. In mixed culture,0.001%? 0.003%?0.005% were all had no significant impact on fatality rate, the height was 28.7% reduced, the blade area was increased by 41%, the density of stomas was reduced by 32.4% and the area of stomas was increased by 58.7% at the concentrate of 0.005%.(4) After 40 d of treatment with three kinds of chemical agents, there were no changes of the number of chromosome in root tip cells which were happened morphological variations were observed, the number of chromosome were all 22.(5) After proliferation of two generations, except the young blade width, the other indicators had no significant impact when compared to control group; the amplification of height was 1.60 cm and 1.58 cm, the blade width was 1.25 cm and 0.88 cm, the blade length was 1.11 cm and 1.12 cm, the density of stomas was 54.37 per mm2 and 54.20 per mm2, the area of stomas was 14842.73?m2 and 14776.29?m2 when soaking with 0.2% Colchicine and mixed culture with 0.05% Colchicine, the indicators were all had significant impact when compared to control group in soaking excepted the width and length of leaf; the amplification of height was 1.19 cm and 0.89 cm, the blade width was 1.18 cm and 0.87 cm, the blade length was 1.16 cm and 0.99 cm, the density of stomas was 50.90 per mm2 and 51.43 per mm2, the area of stomas was 14931.84?m2 and 14801.37?m2 when soaking with 0.2% Pendimethalin and mixed culture with 0.05% Pendimethalin, the indicators were all had significant impact when compared to control group in soaking excepted the width and length of leaf.(6) After the rooting of plants which had clear morphological differences after proliferation of two generations, the rooting was later 5-7 d than control group, the number of root were four to eight less than control group but the roots were stronger and more root hairs.(7) The toxicity of three inducers on the materials was Pendimethalin> Trifluralin> Colchicine. The effects of Pendimethalin were disappeared after proliferation of two generations. As a consequence, the influence of Trifluralin was smaller than Colchicine and Pendimethalin on the growth of tube seedlings.In conclusion, when buds of Pueraria lobata (wild) Ohwi 'Dabashan' mixed culture with 0.05% Colchicine and 0.005% Pendimethalin, its morphological characteristics had significant impact when compared to control group, and roots stronger than control group. It could provide technical reference to breeding of Pueraria lobata (wild) Ohwi 'Dabashan'.
