The optimal tissue culture conditions for callus induction, differentiation, proliferation and rooting of adventitious buds of Pueraria lobata (Willd.) Ohwi were studied through in vitro culture and rapid propagation technologies.The stem segments with axillary buds and leaves from elite parent trees of Pueraria lobata were treated as explants with ethyl alcohol and NaClO according to different concentration gradient and time. The result showed that the optimal sterilization method for the stem segments with axillary buds was 2.2% NaClO for 25-30 min and the survival rate could reach to 50%. Leaf sterilization experiments showed the optimal effect when treated with 2.2% NaClO for 15-18 min and the survival rate could reach to 46%.The results showed that stem segments and leaves of Pueraria lobata could both induct the callus. The most optimal medium for induction of leaf callus was MS+NAA 1.0 mg/L+6-BA 1.5 mg/L, and for stem segment callus was MS+NAA 0.02 mg/L+6-BA 0.4 mg/L+2,4-D 0.4 mg/L. The stem segment callus was better subcultured than leaf callus.The study also showed the most optimal medium for induction of adventitious buds was MS+ NAA 0.1 mg/L+6-BA 0.5 mg/L, and the induction rate reached up to 88.89%. The most optimal medium for differentiation and subculture of the adventitious buds was MS+NAA 0.15 mg/L+6-BA 0.3 mg/L+2,4-D 0.2 mg/L, the differentiation rate reached up to 83.33%, and the multiples of proliferation rate ranged from 6 to 7. The most optimal rooting medium was 1/2 MS supplemented with NAA 0.5 mg/L, and the rooting rate could reach up to 100%.The suitable transplanting medium is a combination of vermiculite:pearlite:peat (in volume)= 6:3:1, and the survival rate of transplantation reached 92%. Overall, a tissue culture system and a relatively good technological system for rapid propagation of Pueraria lobata were established.
