Jujube(Zizjphus jujuba Mill.)is an epibiotic economic fruit tree and an important woody grain species in China,it is rich in nutrients and also of important medical protective value.The fruit cracking of fresh Jujube is the common physiological disease.The fresh fruit is easy get cracking during storage,and it seriously cause the economic loss.In this study,the experiment was carried out in purple planting base of Xinfeng Fruit Industry Co.,Ltd.in Qidong County of Hunan Province.'Mid-Autumn crisp jujube'was used as experimental material.The jujube was soaked with H2O2,Ca(NO3)2,NaCI and ABA.Statistical analysis on the correlation between cracking method,fruit cracking rate,fruit crack index,red index,water absorption,fruit hardness,malondialdehyde,etc.;coomassie brilliant blue was used to determine the soluble protein content in jujube fruit.Kit method determine the water channel protease activity.Real-time PCR was used to determine the expression of aquaporin gene mRNA.And analyzing their relationship with the rate of fruit cracking.The following conclusions were drawn from the research:1.The method of jujube fruit cracking is mainly longitudinal cracking,ring cracking,and the proportion of irregular columns is small.In dark environment,the rate of fruit cracking,the index of fruit cracking and the index of reddening were decreased significantly.Under comprehensive conditions,the treatment of 1%NaCl has thelowest rate of fruit cracking and cracking index,which is the best anti-split treatment.The red index of treatment of 1%NaCl and treatment of 0.25mg/L ABA were higher than the control group..However,in the dark conditions,the index of changing red had a larger fluctuation and was lower than its under the light environment.The lowest water absorption rate is 0.01%NaCl treatment under the light environment.The lowest water absorption rate is 1%NaCl treatment in dark environment.The water absorption rate in the dark environment is generally higher than that in the light environment.The treatment of 90mg/L Ca(NO3)2 had the biggest fruit firmness in the light environment.Only the treatment of 120mg/L Ca(NO3)2 had the higher fruit firmness than the control group.The fruit firmness in light environment was higher than its in dark environment.2.The correlation of different cracking methods is mostly negative,and the correlation between longitudinal crack and ring crack reaches a significant level.Under the light condition,the longitudinal crack+ ring splitting and fruit cracking rate reached a significant level,and the fruit crack index and the red index reached extremely significant levels;the dark environment longitudinal crack + ring crack only reached a very significant level with the cracked fruit index.The fruit cracking rate and the fruit cracking index reached extremely significant levels;the correlation with the reddening index reached a significant level;but it was negatively correlated with fruit firmness and positively correlated with water absorption,and the correlation between the two was not significant.Under the dark condition,the fruit cracking rate and the fruit;cracking index reached a very significant level,and the correlation coefficient was 0.85,which was positively correlated.The rate of fruit cracking was positively correlated with fruit firmness,water absorption and red index,but the correlation was not significant.3.The malondialdehyde content of all treatment groups was higher than that of the control group.Under the light condition,the treatment group of H2O2 had the lowest average malondialdehyde content and there was a negative correlation between malondialdehyde content and fruit cracking rate.Under the dark condition,the treatment group of Ca(NO3)2 had the lowest average malondialdehyde content.But the treatment of 0.005%H2O2 the lowest malondialdehyde content.The content of malondialdehyde was positively correlated with the rate of fruit cracking,and the correlation was not significant.4.Under the light condition,the relative conductivity of the peel and pulp is well coordinated,but in the dark environment,the relative conductivity of the pulp and the peel varies with the concentration of the treatment solution.The perielectric conductivity of all treatment groups was higher than that of the control group.The average conductivity of peel and pulp in the ABA-treated group was lower than that in the other treatment groups.The relative conductivity of the 0.4 mg/L ABA-treated pericarp was the lowest,but it was different from the control group.Insignificant,0.01%H2O2 treated pulp had the lowest relative conductivity,which was significantly different from the control group;in the dark environment,the relative conductivity of all treatment groups was higher than that of the control group,and the relative conductance of the NaCl treated peel was lower than other In the treatment group,the relative conductivity of the peel treated with 0.1%NaCl was the lowest,which was significantly different from the control group.The average relative conductivity of Ca(NO3)2 treated pulp was lower than that of other treatment groups,0.01%NaCl and 0.4 mg/L ABA treatment.The flesh had the lowest relative conductivity and reached a significant level compared to the control.5.The average thickness of the ABA treated stratum corneum was greater than the other treatments.Under light conditions,the rate of fruit cracking was positively correlated with the thickness of the stratum corneum,the thickness of the subcutaneous layer and the thickness of the epidermis,and negatively correlated with the thickness of the epidermis,but they did not reach a significant level.The thickness of the epidermis layer was positively correlated with the thickness of the subcutaneous layer,reaching a very significant level with a correlation coefficient of 0.96.Under dark conditions,the rate of fruit cracking was positively correlated with the thickness of the stratum comeum,the thickness of the subcutaneous layer and the thickness of the epidermis,and negatively correlated with the thickness of the epidermis,but they did not reach a significant level.The correlation coefficient between the thickness of the subcutaneous layer and the thickness of the epidermis was 0.95,reaching a significant level.6.The average soluble protein content of ABA treatment was higher than that of other treatment groups,and 0.1 mg/L ABA treatment had the highest soluble protein content.The water channel protease activity in the light environment is generally lower than that in the dark environment.Under light conditions,Ca(NO3)2 treated water channel protease activity was generally weaker than other treatment groups;in the dark environment,H2O2 treated water channel protease activity was generally lower than other treatments,0.005%H2O2 treated water channel protease The activity was the lowest,reaching a significant level compared to the control,a decrease of 46.2%.In the integrated light and dark environment,the fruit cracking rate was negatively correlated with the water channel protease activity,the correlation coefficient was-0.12;it was positively correlated with soluble protein,and the correlation coefficient was 0.08,but none of them reached significant level.7.The relative expression of aquaporin mRNA in light environment was generally lower than that of aquaporin mRNA in dark environment,and light inhibited the expression of aquaporin mRNA.Under light conditions,the relative expression of ABA-treated aquaporin mRN:A:was generally lower than other treatments.The expression level of water channel protease gene mRNA was negatively correlated with the rate of fruit cracking,but the correlation was not significant.In the dark environment,the relative expression of NaCl-treated aquaporin mRNA was generally lower than that of other treatments.The expression of aquaporin mRNA was positively correlated with the rate of fruit cracking,but the correlation was not significant.
