Populus nigra cv.'Jinye' is a variety of Populus nigra,Populus,Its biological characteristics are similar to P.nigra.In this experiment,we explored the axillary bud culture induced by stem,adventitious bud culture and induced callus induction by leaf,rooting culture,seedling acclimatization and management of P.nigra cv.tJinye,.The results were as followed:1.Axillary bud culture induced by stem of.nigra cv.'Jinye'.The results showed that the best disinfection method for annual stems was 0.1%HgCl2 for 10 min.The infection rate of stem was 10%and the survival rate was 77.3%.For axillary bud induction using stem,the highest induction frequency of axillary bud was 92.6%,using MS containing 0.5 mg/L 6-BA and 0.3 mg/L NAA.The best hormone ratio for proliferation was 1.0 mg/L 6-BA and 0.4 mg/L NAA,additional 0.2 mg/L GA3.On this medium,the multiplication coefficient can reach to 4.4.2.In the adventitious bud induction pathway using leaf of P.nigra cv.'Jinye'.The optimum disinfection method of tender leaves was using 2%NaCIO for 9 min,and the survival rate was 76.7%.MS medium containing 1.0 mg/L 6-BA and 0.3 mg/L IBA given the highest rate of adventitious bud induction,the value was 95.5%,and the average number of induced buds was 19.9.The best hormone ratio for proliferation was on MS with 1.0 mg/L 6-BA,0.4 mg/L IBA and GA3 0.2 mg/L,the multiplication coefficient was 4.53.3.Using leaf to induct the callus of P.nigra cv.'Jinye',the results showed the MS medium with 1.0 mg/L 6-BA and 0.6 mg/L 2,4-D has great induction effect in dark,and the induction rate was 85.8%.The best callus proliferation was on MS medium with 1.5 mg/L 6-BA and 0.6 mg/L NAA,and the multiplication coefficient was 4.47.Optimum hormone ratio for callus differentiation culture was 1.0 mg/L 6-BA and 0.1 mg/L IBA,the differentiation rate reached 84.3%,and the average number of differentiation buds was 22.2.4.For seedling rooting of P.nigra cv.'Jinye',at the same concentration,the induction effect of IBA was better than NAA.The roots induced by IBA were strong and long,more fibrous roots and higher rooting rate.While NAA induced roots,the main roots were strong,but it had fewer fibrous roots and the roots were brittle and fragile.So,we inducted seedling rooting on 1/2 MS and 0.5 mg/L IBA,the rooting rate was 91.2%.5.In the process of seedling acclimatization and management of P.nigra cv.'Jinye'.The results showed that bottle with seedlings should be opened for 2 days to make the seedlings adapt to the environment of the nursery gradually.Then taked out it and planted in soil matrix with four to two to one of humus,vermiculite and perlite,the highest survival rate was 81.1%.During the daily management,set the light intensity about 4000 lx and the humidity 60%?70%,and sprayed the nutrient solution properly to promote its growth.By comparing the three culture methods,it was found that stem-segment-induced axillary bud culture could be the best way for tissue culture of P.nigra cv.'Jinye'.
