| The study was conducted to investigate the effect of dietary supplementation with coated cinnamon oil as an antibiotic substitute on the immunity function and the expression of intestinal mucosal barrier gene of broiler chickens.In a single-factor designed trial,a total of 396 one-day-old Cobb broiler chicks were randomly assigned into 6 groups,with six replicates per group,and each replicate have 11 birds.Birds in the control group were fed the corn-soybean meal basal diet,and birds in the positive control were fed basal diet supplemented with 50mg/kg of flavomycin.In other four groups,birds were fed basal diet supplemented with 50,100,200 and 300 mg/kg of coated-cinnamon,and we marked these experimental groups as experimental groupⅠtoⅣ.The experimental breeding period is 42 days.The results were showed as follows:(1)Effects of dietary supplementation with coated cinnamon oil on growth performance and immune organ index of broilers.Compared with the control group,dietary supplementation with coated cinnamon oil had no significant influences on average daily gain(ADG),average daily intake(ADFI),thymus index,spleen index and bursa of Fabricius index(P>0.05).(2)Effects of dietary supplementation with coated cinnamon oil on peripheral blood immune function of broilers.Compared with the control group,the content of lysozyme in serum of 21 day and 42 day old broilers were up-regulated by the coated cinnamon oil treatment group(P<0.05),and with the increase of the dosage of coated cinnamon oil in the diet,the content of lysozyme in serum were increased linearly(P<0.001)and curvilinearly(P<0.001).And also,it had a linear positive correlation.The content of lysozyme in serum of 21 day old broilers showed linear positive correlation with the increase of the dosage of cinnamon oil added in the diet(R~2=0.824,n=30);the result of the 42 day old broilers is the same as before(R~2=0.625,n=30).The addition of cinnamon oil in the diet had no significant effect on the titer of NDV antibody in the serum of 21 day old broilers(P>0.05).Dietary with 200 mg/kg coated cinnamon oil significantly decreased the NDV antibody titer in the serum of 35 day old broilers(P=0.005).(3)Effects of dietary supplementation with coated cinnamon oil on the content of Immunoglobulin of broilers.At the age of 21 days,compared with the control group,the 50 mg/kg coated cinnamon oil increased the content of serum immunoglobulin A(IgA),and reduced the content of serum immunoglobulin G(IgG),and also reduced the content of secretory immunoglobulin A(sIgA)in the jejunum and ileum mucosa(P<0.05).The 100 mg/kg coated cinnamon oil reduced the content of sIgA in the ileum mucosa(P<0.05).The 200 mg/kg coated cinnamon oil increased the content of serum IgA and IgG,and reduced the content of sIgA in the ileum mucosa(P<0.05).The 300 mg/kg coated cinnamon oil increased the content of serum IgG and reduced the content of sIgA in the ileum mucosa(P<0.05).Compared with the positive control group,the 50 mg/kg coated cinnamon oil increased the content of serum IgA and the content of sIgA in the duodenum and jejunum mucosa,but it also reduced the content of sIgA in the ileum mucosa(P<0.05).The 100 mg/kg coated cinnamon oil increased the content of serum IgA,IgG and the content of sIgA in the jejunum mucosa(P<0.05).The200 mg/kg coated cinnamon oil increased the content of serum IgA,IgG and the content of sIgA in the duodenum and jejunum mucosa(P<0.05).The 300 mg/kg coated cinnamon oil increased the content of serum IgA,IgG and the content of sIgA in the jejunum mucosa(P<0.05),too.At the age of 42 days,compared with the control group,dietary with 50 mg/kg coated cinnamon oil reduced the content of serum IgG(P<0.05).Dietary with 100 mg/kg coated cinnamon oil increased the content of sIgA in the jejunum mucosa,but reduced the content of serum IgA,IgG and the content of sIgA in the jejunum mucosa(P<0.05)Dietary with 200 mg/kg coated cinnamon oil reduced the content of serum IgG and sIgA in the ileum mucosa(P<0.05).Dietary with 300 mg/kg coated cinnamon oil increased the content of sIgA in the jejunum mucosa and reduced the content of serum IgA(P<0.05).Compared with the positive control group,all dietary supplementation with coated cinnamon oil group increased the content of sIgA in the jejunum mucosa and reduced the content of serum IgA(P<0.05).The experimental group which dietary with 50,100,200 mg/kg coated cinnamon oil reduced the content of serum IgG,and the 300 mg/kg coated cinnamon oil increased the content of serum IgG(P<0.05).(4)Effects of dietary supplementation with coated cinnamon oil on T,B lymphocyte transformation rate in peripheral blood of broilers.At the age of 42days,Adding coated cinnamon oil to the basal diet decreased the ConA stimulation index of peripheral blood lymphocytes(P=0.003),and increased the LPS stimulation index(P<0.001).Compared with the control group,dietary with 100and 300 mg/kg coated cinnamon oil have a significant reduction in the ConA stimulus index,however,all coated cinnamon oil had a significant increasion in the LPS stimulus index(P<0.05).This result is the same as these which was compared with the positive control group.(5)Effects of dietary supplementation with coated cinnamon oil on the mRNA expression level of cell immune factor in intestinal mucosa of broilers.At the age of 21 days,compared with the control group,the 50 mg/kg coated cinnamon oil up-regulation the mRNA expression level of IL-6 in duodenum(P<0.05),the 50,100,300 mg/kg coated cinnamon oil up–regulation the mRNA expression level of IL-8 in ileum(P<0.05),the 200 mg/kg coated cinnamon oil up–regulation the mRNA expression level of IL-1βand IL-8 in jejunum(P<0.05).Compared with the positive control group,the 50 mg/kg coated cinnamon oil up-regulation the mRNA expression level of IL-6 in duodenal and IL-8 in ileum(P<0.05).the 100 mg/kg coated cinnamon oil up–regulation the mRNA expression level of IL-6 and IL-8 in ileum(P<0.05),the 200 mg/kg coated cinnamon oil up–regulation the mRNA expression level of IL-1βand IL-8 in jejunum(P<0.05),the 300 mg/kg coated cinnamon oil up–regulation the mRNA expression level of IL-8 in ileum(P<0.05).At the age of 42 days,compared with the control group,all dietary with coated-cinnamon groups down-regulation the mRNA expression level of IL-8 in duodenum(P<0.05).And the 200,300 mg/kg coated cinnamon oil down-regulation the mRNA expression level of IL-8 in ileum(P<0.05),the 300 mg/kg coated cinnamon oil down-regulation the mRNA expression level of IL-1βin jejunum(P<0.05).Compared with the positive control group,the 100 mg/kg coated cinnamon oil up-regulation the mRNA expression level of IL-1βin ileum(P<0.05),and down-regulation the mRNA expression level of IL-6 in ileum(P<0.05);the200 mg/kg coated cinnamon oil up-regulation the mRNA expression level of IL-1βin duodenum(P<0.05),and down-regulation the mRNA expression level of IL-6in ileum(P<0.05);the 300 mg/kg coated cinnamon oil up–regulation the mRNA expression level of IL-1βand IL-8 in duodenum(P<0.05).(6)Effects of dietary supplementation with coated cinnamon oil on intestinal barrier function of broilers.Compared with the control group,the mRNA expression level of Occludin in the jejunum mucosa of 21 day old broilers up-regulated by the experimental group of coated cinnamon oil(P<0.05).Dietary supplementation with 200 mg/kg coated cinnamon oil also increased the mRNA expression level of Occludin in the ileum mucosa of 42 day old broilers(P<0.05).300 mg/kg coated cinnamon oil increased the mRNA expression level of Claudin-1in the intestinal mucosa of both 21 and 42 day old broilers(P<0.05).50 mg/kg coated cinnamon oil up-regulation the protein expression of Claudin-1 in the ileum mucosa of 42 day old broilers(P<0.05).Conclusion:Under this experimental condition,dietary supplementation with50 mg/kg coated cinnamon oil can improve the immune function of 21 day old broilers. |
