AIM:Fatty liver hemorrhage syndrome(FLHS)is a common nutritional metabolic disease in laying hens due to the disorder of fat metabolism in laying hens and excessive fat deposition in the liver,which leads to steatosis,hemorrhage and necrosis of liver cells,resulting in the decline of egg production rate in the population and even death.Previous studies of our research group have found that taurine can regulate fat metabolism,improve fat deposition and reduce liver steatosis in fatty liver rats.Meanwhile,taurine can also reduce mitochondrial oxidative stress in liver cells,protect mitochondrial function and prevent FLHS,but the specific mechanism is still unclear.The purpose of this study was to establish a FLHS model of laying hens,add taurine into the diet,detect the effect of taurine on the production performance and quality of FLHS laying hens,and explore the effect of taurine on the liver mitochondrial biosynthesis and autophagy of FLHS laying hens,so as to provide theoretical basis for the application of taurine to prevent the occurrence of FLHS in laying hens.Methods:select 300 only 52 weeks of highland brown laying hens,randomly divided into 6 groups: normal control group(C),taurine group ?(T1),taurine group ?(T2),FLHS model groups(M),taurine ? prevention group(MT1),taurine ? prevention group(MT2).Laying hens in groups C,T1 and T2 were fed soy-corn-type basal diets,while laying hens in groups M,MT1 and MT2 were fed high-fat and low-protein diets.Taurine(0.05%)and taurine(0.3%)were added to diets in groups T1,MT1,T2 and MT2 for 16 weeks.During the experiment,eggs were collected and weighed to detect the production performance and egg quality of laying hens.At the end of the experiment,liver was collected and total liver RNA was extracted.q RT-PCR was used to detect mRNA expression of mitochondrial biosynthesis and related indicators of liver autophagy.Results: 1.Compared with the normal control group,the egg-laying rate in the FLHS model group was significantly reduced,and the egg-laying ratio was significantly increased(p<0.01).Eggshell strength,eggshell thickness,protein height,haversian unit and yolk color all decreased significantly(p<0.01).mRNA expression levels of liver mitochondrial biosynthesis marker genes PGC-1?,NRF1,NRF2 and TFAM were significantly decreased(p<0.05 or p<0.01).Liver cell autophagy related genes LC3?,LC3?,Beclin1 mRNA expression level decreased significantly(p<0.05 or p<0.01),P62 mRNA expression level significantly increased(p<0.01);2.Compared with the model group,taurine prevention group had significantly higher egg production rate and significantly lower egg production ratio(p<0.01).Eggshell strength,eggshell thickness,protein height,haversian unit and yolk color were significantly increased(p<0.05 or p<0.01).mRNA expression levels of PGC-1?,NRF1,NRF2 and TFAM were significantly increased(p<0.05 or p<0.01).LC3?,LC3?,Beclin1 mRNA expression level was significantly increased(p<0.05 or p<0.01),P62 mRNA expression levels significantly lower(p<0.01).In conclusion,taurine can improve the production performance and egg quality of FLHS laying hens,increase the mitochondrial biosynthesis of hepatocytes,improve the autophagy inhibition of hepatocytes,and play a role in preventing FLHS of laying hens.
