Coenurosis is a serious zoonotic parasitic disease caused by the metacestode(Coenurus cerebralis)of Taenia multiceps,which parasitic on the central nervous system of humans and animals.This disease is a worldwide distribution,which is more commonly in the countries and regions dominated by herbivorous animals,especially in the areas such as the north,northwest,and the northeast of China.The incidence of the goats in the local areas such as Gansu,Ningxia,Qinghai and Xinjiang can reach 53%.Coenurosis often affects the health of herbivores,resulting in great economic loss to animal husbandry.Therefore,effective prevention and accurate diagnosis are the important steps to control the disease.In this study,the biological functions of Tm-HSP60 and Tm-p36 of T.multiceps were investigated,the potential targets for the prevention and treatment of coenurosis were selected,and provided important reference data for the study of T.multiceps.At the same time,the serological diagnostic potential of Tm-Ag B and Tm-TPx and Tm-HSP60 in diagnosis of coenurosis were evaluated,and the most promising antigen was selected.And this study provided a reliable technical support for accurate diagnosis of this disease.1 Molecular characterization and expression analysis of two heat-shock proteins in Taenia multicepsIn this study,we performed sequence,structure,tissue distribution and transcriptional profile analyses of two heat-shock proteins(HSPs)in T.multiceps and found that both of them share typical highly conserved features of HSPs.Tm-p36 shares structural characteristics with metazoan small HSPs,with two N-terminal α-crystallin domains.The result indicates that they may play important roles in the life-cycle of T.multiceps as molecular chaperones.Tm-HSP60 is widely distributed in all life-cycle stages,and compared with Tm-p36,Tm-HSP60 displayed stronger immunoreactivity.Thus,Tm-HSP60 is of interest for antibody detection,while Tm-p36 was most strongly associated with oncospheres and protoscolices,and is of potential interest for vaccination for controlling C.cerebralis in the future.2 Expression of Ag B,TPx of Taenia multiceps and evaluation of its diagnostic potentialIn this study,recombination protein Tm-Ag B and Tm-TPx was expressed,and the Tm-Ag B sequence was analyzed by bioinformatics.The results showed that Tm-Ag B was homologous to Ag B5 of Echinococcus granulosus and Echinococcus multilocularis.We also evaluated the serological diagnostic potential of four antigens,r Tm-HSP60,r Tm-Ag B,r Tm-TPx and r Tm-Anx B2 to establish indirect ELISA,and compared them with the previously reported r Tm-HSP70 and r Tm-GST.The results showed that r Tm-Ag B with a sensitivity of 95.8% and a specificity of 87.5% have a stronger potential for serological diagnosis,we used it as a diagnostic tool to investigate the infection in the endemic area of coenurosis in China,and found that 18 of the 100 sera were suspected to be infected.In this study,a potential antigen for serological diagnosis was screened,which provides a reliable tool for the diagnosis of T.multiceps in the future.
