Gathered the Coenurus cerebralis,ultrathin section and tissue slice of scolex,capsule walland cervical part of the Coenurus cerebralis were observed by transmission electron microscope(TEM) and optical microscope.To observe the mature Coenurus cerebralis by optical microsope, Paraffin method and HEmethod were used.The results indicated that the outer part of the scolex is very different from theouter part of the capsule wall; The musle which composed the holdfast appear radialdistribution,there are some vacuoles and particles can be seen in stripe shape organization, thereare some microtrichia in its surface,the scolex can be divided into holdfast,rostellum,somatoystand body-cavity according to the abvious differences.There are two layers on somatoyst,the outeris corneum,the inner has nearly200scolex is germinal layer.Ultrathin section of scolex,capsule wall and cervical part of the Coenurus cerebralis wereobserved by transmission electron microscope (TEM).The results indicated that cyst wall can bedivided into tegument,matrix and parenchyma.Tegument cell,parenchyma cell,calcareouscorpuscle,myoblast,flame cell and emunctory system can be seen on the parenchyma region.Thestructure of well developed muscle fiber can be found on scolex,inside the scolex there arestructures similar to the tegument on capsule wall,and thick microtriche.the kinds of celldecrease and the number of veses which contain granula increase.The indication can beconcluded that the structure of capsule wall and variety of cells from Coenurus cerebralis aresimilar to other tapeworms,and the emunctory system of Coenurus cerebralis much moreabundant.Structure of scolex and cervical part are different from other tapeworms.Gather taenia multiceps from sheep which naturely infected oncosphere,breed dogs for theanimal model,gather gravid segments,eggshell was destroyed by sodium hypochlorite,scoleswere actived by simulated intestinal fluid,design Special primer, RT-PCR was used to amplifythe Tm18gene from Taenia multiceps, Cloning into the pGEX-4T-1vector,transform intoBL21,express recombinant protein guided by IPTG,the product analysis by SDS-PAGE andWestern-blot,the result indicate that recombinant protein had strong reactivity with sera ofnaturely infected sheep,and can be used for the candidate vaccine antigen.
