Research Background:Ketotoxicity is a common nutritional metabolic disease in perinatal dairy cows.Its main pathological features are hyperketosis,high non-esterified fatty acids(NEFAs)and hypoglycemia.Peroxisome proliferator-activated receptors(PPARs)are a family of nuclear receptors composed of PPAR?,PPAR?/? and PPAR?,which are responsible for regulating cell differentiation and lipid metabolism,and are involved in the regulation of various biological functions in vivo..Research purposes:In this study,the effects of BHBA and NEFAs on the expression of peroxisome proliferator-activated receptor gene were detected by adding exogenous ?-hydroxybutyrate(BHBA)and NEFAs to bovine endometrial cell culture medium cultured in vitro.To provide a theoretical basis for further research on the effects of ketosis on the metabolism of dairy cows and its relationship with other common diseases in perinatal period.Research method:In this experiment,bovine endometrial cells(BEND)were cultured in vitro,and concentrations of 0,0.6,1.2,2.4 mmol/L BHBA and 0,0.3,0.6,1.2 mmol/L NEFAs were added to extract total cellular RNA and The total protein,and then the mRNA and protein expression levels of PPAR?,PPAR? and PPAR? in BEND cells were detected by real-time fluorescent quantitative PCR and immunoblotting.The effects of different concentrations of BHBA and NEFAs on the expression of PPARs were studied.Result:Compared with the control group,exogenous BHBA could inhibit the expression of PPAR? and PPAR? at BHBA concentration of 0.6 and 1.2 mmol/L,inhibit the expression of PPAR? at 0.6 mmol/L,and promote the expression of PPAR? and PPAR? at BHBA concentration of 2.4 mmol/L.With the increase of BHBA concentration,the protein expression of PPAR?,PPAR? and PPAR? also increased significantly.Compared with the control group,exogenous NEFAs could significantly promote the expression of PPAR? in BEND cells in a dose-dependent manner,significantly inhibit the expression of PPAR?,but not in a dose-dependent manner,while different concentrations of NEFAs had no significant effect on the expression of PPAR? in BEND cells.With the increase of NEFAs concentration,the protein expression of PPAR? also increased significantly;exogenous NEFAs could significantly increase the protein expression of PPAR? at the concentration of 0.3 and 1.2 mmol/L,and decrease the protein expression of PPAR? at the concentration of 0.6 mmol/L.Conclusion:High concentrations of BHBA and NEFAs can alter the expression levels of PPAR?,PPAR? and PPAR? in bovine endometrial cells,and may further affect various metabolic processes in vivo through peroxisome proliferator-activated receptors(PPARs).
