Acariasis is a parasitic disease that is chronic,contact,and common on animal body.During today's farming industry,it is the most common,especially Psoroptidae and Sarcoptidae.Chemical drugs are widely used to treat mite.However,The high residual residual,resistance and environmental hazards have been caused by chemical drugs,and the whole society had also paied close attention to that.Eupatorium adenophorumbelong to perennial herb in Mexico,which native to the Americas.When people studyingdeeply on this invasive plant,people had found that it had bring ecological hazards to more than 30 countries.But people also had found that chemical substances,extracted from theEupatorium adenophorum,can kill many agricultural pests and animal parasites,especially inPsoroptidae and Sarcoptidae.So it has great potential to be a botanical acaricide.To develop new drugs for the treatment of mites,we had studied the extraction process ofEupatorium adenophorum.In this study,we had deal withthe extracted liquid,obtained from Eupatorium adenophorum,and had assessed its acaricidal effect,toxicology,and safety in vitro.Results are as follows:1.Study on extraction of ethanol from Eupatorium adenophorumSingle factor and orthogonal test was adopted.Using high performance liquid chromatography,9-OXO-10 and 11-dehydro-ageraphorone,which is the main acaricidal active ingredient,was chosen to be the detective index.The orthogonal test was carried out to investigate the influence of volume percentage of ethanol?solid to liquid ratio?extraction duration and extraction times on the extraction progress.We designed L9?34?orthogonal experiment to determine the optimum extraction conditions.The result:95%volume percentage of ethanol,solid to liquid ratio of1:20,extraction duration of 1 h,extraction times of 3 h.2.Evaluation of the safety and the acaricidal effect of Eupatorium adenophorum in clinical by transdermallyAcute toxicity and irritation test of skin was carried.The results had shown that:there was no acute toxicity reactionon the animals.According to the standard for evaluation of skin irritatin,there was noskin irritation on animalsafter 72 hours after complete skin and damaged skin test.Acaricidal test in clinical:rabbits were randomly divided into six groups.Group A and B were treated with 1.0g·mL-1?0.5g·mL-1 drug which containingAzone,respectively.Group C and D were treated with 1.0g·mL-1?0.5g·mL-1 drug which withoutAzone,respectively.Group Ewere treated with 0.2 mgIvermectinby injecting?positive controls?.GroupF were treated with saline by embrocating?negative controls?.The results showed that,the high-dose groupwith Azone worked best in psoroptesmites group and scabies mitesgroup.3.Study on acute toxicity test of Eupatorium adenophorumtransdermal agentThe KM mice were divided into different groups.The preliminary experiment was carried out repeatedly to determine the dose range.Sixty KM mice were testedby orally administration in formal experiment.We observed and recorded the toxic reactions and the death of the mice 7 d,consecutively.LD500 and 95%confidence limit were calculated by using Karber metod.The results showed that:LD50=92.68g/kg,95%confidence limit was 87.02105.15g/kg.The transdermal agentcan makelesion in heart,liver,spleen,lung,kidney,and intestine.4.Subchronic toxicity test ofEupatorium adenophorumtransdermal agentEighty SD rats were randomly divided into 4 groups?each 20 rats per group?.The rats were treated with different doses by orally for 28 dconsecutively:high-dose group(1/10LD50),intermediate-dose group(1/25LD50),low-dose group(1/50LD50)and 0g/kgEupatorium adenophorumtransdermal agent.We had detected and recorded the clinicalmanifestations,weight change,hematological indexes,gross anatomy,organ index and histopathology in organs.The results showed that Eupatorium adenophorumtransdermal agent had different effects on growth,hematology and organ index in sex.But it had no regularityon...
