Piglet diarrhea as a common clinical disease,is a serious threat to the steady and healthy development of pig industry.Due to the acute onset,diverse pathogens,complicated infections and the lack of accurate pathogen diagnosis in the course of clinical treatment,the disease can not been controlled rapidly and effectively,causing incalculable economic losses to pig industry.Due to different climatic and feeding conditions in different regions,the incidence of diarrhea in piglets and the main pathogens are consequently different.Therefore,this paper combines the microbiology detection-bacterial culture isolation and purification with molecular biology-16 SrDNA Identification to investigate the major pathogens from piglet diarrhea in Jilin from March 2017 to March 2018.One hundred and seventeen samples of diarrhea were tested during this period.Microbiological tests revealed 104 samples of suspected E.coli causing diarrhea and 19 samples of suspected Salmonella-causing diarrhea.Subsequently,the pathogenicity test of the isolated and purified bacterial liquid was carried out.The results showed that 49 of 104 samples were pathogenic and 15 of 19 samples were pathogenic.Synthetic 16 SrDNA universal primers and specific primers for pathogenicity of the samples were identified the results showed that 49 samples were Escherichia coli,accounting for 42%(49/117).All 15 samples were Salmonella,accounting for 13% of the total(15/117).The results showed that the main pathogen of bacterial diarrhea in piglets in Jilin area was Escherichia coli.Three samples were selected for target gene recovery,target gene-vector connection,gene transformation and selection and sequencing after recombinant plasmids.Then,we compare the sequencing results with the E.coli gene sequence published in GenBank and found that its homology was up to 99% with no mutation.The results of susceptibility test of 49 diarrhea piglets caused by pathogenic E.coli showed that the pathogens in some areas of Jilin City were highly sensitive to levofloxacin,enrofloxacin,ciprofloxacin hydrochloride,amikacin sulfate,Clindamycin and norfloxacin,moderately sensitive to neomycin sulfate,gentamicin,diarrhea net and azithromycin and highly resistant to amoxicillin,ceftriaxone sodium,clarithromycin,sulfamonomethoxine,tylosin tartrate,apramycin sulfate,timustacin and lincomycin hydrochloride.
