Anti-mullerian hormone(AMH)can be used as a reliable indicator to evaluate the function of goat's ovarian reserve.The study found that there are differences in the AMH gene polymorphism in different races,and AMH gene polymorphism affects the ovarian reserve function.Then,in livestock breeding,is there any difference in goat's ovarian reserve function caused by different AMH gene polymorphisms in different goat breeds,resulting in differences in reproductive performance? At present,there is no report about the effect of molecular level variation of goat AMH gene on ovarian function reserve.In this experiment,four goat breeds such as Dazu Black Goat,Inner Mongolian Cashmere Goat,Boer Goat,and Nub Goat were sampled.Firstly,by using PCR-SSCP,AMH single nucleotide polymorphisms were detected and analyzed for AMH single nucleotide polymorphisms in a total of 200 individuals of 4 goat breeds(50 for each breed)to determine whether there was a difference in AMH gene polymorphisms.Then,heart,liver,spleen,lung,kidney and ovary of Dazu black goat,Inner Mongolia cashmere goat and Boer goat were collected.The expression of AMH in each organ was analyzed by RT-PCR.Finally,4 goat breeds were subjected to blood sampling from the jugular vein.AMH ELISA kits were used to measure AMH levels of in blood among different species,and the one among same species(Dazuo black goat)on 9th,15 th,and 31 th days,6 months,1 year,2 years,and 3 years of age.The molecular mechanism of goat ovarian reserve function provides an important theoretical basis and data support.The results of this experiment based on this study are as follows:1.SSCP analysis showed that there was no polymorphism in the AMH gene and its receptor gene AMHR2 between the four breeds of Dazu black goat,Inner Mongolia cashmere goat,Boer goat and Nubian goat.2.Sequencing the PCR products for rs662076763,rs649255341,rs647977151,rs663303947,rs672197988,and rs650783324 found on NCBI.The experimental results showed no mutations in the above sites in each of the 25 samples.3.The AMH gene in goat is expressed only in ovary and is not expressed in other organs.There was no significant difference in AMH m RNA among Dazu black goat,Inner Mongolian cashmere goat,Boer goat,and Nub goat.4.During the lactation period,there was no difference in the AMH level between the blood of Dazu black goat and Nub goat(P>0.05),and there was no difference in the AMH level between Boer goat and Inner Mongolian cashmere goat(P>0.05),but the AMH level in Dazu black goat and Nub goat had extremely significant differences with the AMH level in Boer goat and Inner Mongolia cashmere goat(P<0.01).At the age of 6 months,the AMH levels in the blood of Dazu black goat and Inner Mongolia Cashmere Goat were not significantly different(P>0.05),but Boer goat were 12.6%(P<0.05)and 36.7%(P<0.01)higher than the AMH levels of Dazu goat and Nub goat,respectively.In adulthood,there was no significant difference in blood AMH between Dazu Black Goat and Boer Goat,Nobi Goat or Inner Mongolia Cashmere Goat(P>0.05),but the AMH level of Boer Goat and Nub Goat was higher than that of Inner Mongolia Cashmere Goat 13.5% and 15.9%,respectively(P<0.05).5.In Dazu black goat,the level of AMH hormone gradually decreased with age.The AMH levels at 9 days of age were 13.9%,15.2%,29.4%,and 32.1% higher than those at 6 months,1 year,2 years,and 3 years,respectively(P<0.01).
