Artificial insemination of Muscovy ducks has become one of the important means of reproductive management and plays an important role in production practice.Through semen preservation technology,it is possible to better promote the dissemination and sharing of excellent genetic resources,prolong the service life of superior male Muscovy ducks,reduce breeding costs,and improve production efficiency.The low-temperature liquid preservation technology of Muscovy duck semen directly determines the quality of semen preservation.In this experiment,the semen of Muscovy duck was used as the research object.Firstly,four diluents(YA-1,YA-2,YA-3,and NO-1)were prepared,and the Muscovy duck semen was diluted in the four diluents in ratios of 2:1,1:1,and 1:2,respectively.The diluted semen was then stored in a 4 ℃ environment for four consecutive days.The effects of different diluents and dilution conditions on the sperm motility and other apparent parameters during low-temperature liquid preservation of semen were investigated,and the related mechanisms were explored.The ultimate goal was to develop an effective Muscovy duck low-temperature preservation diluent formula that could further improve sperm quality and establish a low-temperature liquid preservation technology system for Muscovy duck semen.The research results are as follows:(1)Different diluents have a significant impact on the vitality,motility,and other motion-related parameters of the sperm.Among them,the YA-1 diluent demonstrated the best preservation effect on sperm,with all the motion-related parameters being significantly higher than those of the sperm preserved in other diluents on the 3rd and4 th days(P<0.05).(2)When choosing the dilution ratio under the YA-1 diluent,if the semen preservation time is less than 48 hours,the 1:1 dilution ratio is the best for protecting the sperm;if the preservation time is over 48 hours,the 1:2 dilution ratio is the best for protecting the sperm.(3)In the testing of sperm reactive oxygen species(ROS)and acrosome integrity,as the preservation time of the sperm increased,the protective effect of YA-1 diluent on acrosome integrity was most pronounced,and it was able to reduce ROS production.(4)In the artificial insemination experiment,the highest hatching rate was observed in the sperm mixed with YA-1 diluent,with over 25% hatching rate even after 3 days of preservation.This indicates that YA-1 diluent has obvious advantages in protecting sperm and promoting reproductive capacity.
