The ancient trees are "living fossils" carrying Chinese civilization,which contain strong historical and cultural connotations.The ancient ginkgo trees are respected as the"national trees",symbolizing the traditional spirit of perseverance and tenacity of the Chinese nation,and they are valuable materials for studying Chinese culture.At the same time,after thousands of years,ancient trees still maintain tenacious vitality,which is closely related to their excellent genetic genes.An ancient tree is a valuable gene pool,so it is great significance to study the propagation of ancient trees.Due to the change of external environment,human destruction and other factors,the survival of ancient trees has been greatly threatened,coupled with the decline of the physiological mechanism of ancient trees,the reproductive capacity of ancient trees has gradually decreased,so it is urgent to improve the propagate technology of ancient tree.This paper takes ancient ginkgo biloba,which grows in Louguantai,Zhouzhi county as an example.Through the research on their seed germination treatment technology,pollen preservation treatment technology,cutting treatment method and callus induction technology to explore the techniques suitable for seed breeding,pollen preservation,cutting propagation and callus induction on ancient ginkgo,so as to provide technical support for the propagation of ancient ginkgo.The result is as follows:1.The condition of changing temperature?first 35?for 2 days and then 26?for continuous germination?is more beneficial for ancient ginkgo biloba seeds germinate rapidly and neatly than the other two.The best treatment for ancient ginkgo biloba seeds to germinate is to accelerate the germination at the changing temperature condition?35?for 2days,then continue to germinate at 26??;The damage cause scale of ancient ginkgo biloba seeds from high to low are as follows:withered,worm-eaten,bacterial infection and mechanical damage,according to different conditions,targeted solutions should be taken.2.The pollen of ginkgo biloba was stored at room temperature/4?/-30?.The results show the condition of 4?is the most beneficial for ginkgo biloba pollen to keep its activity,and in this condition,the pollen could be used in the production of ginkgo biloba within the first 30 days.The pollen activity in-30?cryogenic refrigerator decreased the fastest,which is the least suitable for the preservation of ancient ginkgo biloba pollen.3.The most suitable cutting treatment for semi-woody branches of ginkgo biloba is that200mg/L ABT-1 soaking for 5 minutes,and the survival rate is 76.67%.4.The healthy and young leaves of ginkgo biloba were used as tissue culture materials,9 medium formulas were set up,inoculated in aseptic environment,cultured for 30 days,and the color and morphological changes of callus were observed and compared.The results show that the most favorable medium for callus induction of ginkgo biloba leaves is?7?MS NAA3.0mg·L-1+KT0.1mg·L-1+sucrose 30g·L-1+plant gel 2.25mg·L-1.
