Ips spp.belonging to the Coleoptera,Scolytidae,Ipinae,has more than 60 species in the world,18 species belong to quarantine pests and 25 are native to North America.Ips is considered to be one of the most destructive attacking beetles.If some species of this genus come to China,it may cause serious damage.In North America,Canada is also China's main log import area.Therefore,we should pay more attention to quarantine inspection of imported timber from Canada.The accurate identification of harmful organisms is the precondition of quarantine.In addition,the adult morphology of the genus beetle is very similar,the identifiable features are limited,the individual differences exist and the port interception is easy to cause the insect's limb damage,which affects the accuracy of the morphological identification.In recent years,molecular identification technology has developed rapidly.So in this paper,the DNA bar code technology is used to study the genus beetle from the Canadian spruce,so as to make a direct and effective identification of the Ips captured on the Canadian spruce logs at the port.The main results are as follows:1.we studied the morphological classification of the Ips,analyzed the identification and identification characteristics of the Ips,and acquired various kinds of high-definition maps of the back,abdomen and side.2.we extracted genomic DNA and screened the suitable PCR amplification system to amplify the COI gene sequence.By analyzing the amplified sequences and designing primers for the genus Picea beetle,5 species of Ips were amplified successfully,indicating the generality and feasibility of the primers to amplify the genome DNA of the genus Canadia beetle.3.COI gene sequence analysis.The COI gene amplification sequence of Ips was collected and corrected.The genetic distance and phylogenetic relationship were calculated by MEGA.The final results show that the intraspecific and intermediate distance conforms to the standard of bar code,and it is suitable for the identification of the genus beetle of the Canadian spruce.4.DNA bar code is initially built.Based on the genetic distance and phylogenetic tree,the COI gene sequence of the genus beetle was analyzed and evaluated.It was found to be in line with the bar code test standard and can be used for species identification.The DNA barcode of this genus was preliminarily constructed.The completion of the development of Canada spruce DNA barcoding technology research.
