Pyometra is one of the most common reproductive diseases in bitches and female cats.Bacterial infection and reproductive hormone disturbance are the main causes of pyometra.At present,the liquid state in the uterus is treated as pyometra,but according to the existing research,most of the uterine lesions do not develop to pyometra stage.Due to the limitations of diagnosis,the method of hysterectomy is adopted for the above-mentioned cases,which is harmful to pets.In this study,109 cases of pyometra were collected from the outpatient department of some animal hospitals in Harbin,including 90 bitches and 19 female cats.Through statistical analysis of the basic situation,clinical symptoms,laboratory examination and other results,the collected uterine tissues were made into paraffin sections,stained with hematoxylin-eosin(HE)and observed according to the histopathological results.At the same time,real-time quantitative polymerase chain reaction(q PCR)was used to detect the microbe in the uterine contents of bitches and female cats after the onset of the disease.Summarizing the regularity of pyosis accumulation in Harbin area,and provided the basis for clinical diagnosis and treatment of bitches and f emale cats pyometra.The results showed that the average age of onset was 7.50 ± 3.13 years old,the age range was 1.5-13 years old,and the most affected dogs are over 6 years old.Among them,the Miniature poodle(31.11%)and Chinese rural dog(13.33%)were the susceptible breeds,and the incidence was the most in 2-6 months after oestrus;The average age of feline onset was 6.74 ± 3.64 years old,and the onset age ranged from 1 to 14 years old,and the incidence of multiple cats over 6 years old(68.42%),of which the Dragon Li(30.7%)and the British Shorthair(23.08%)were susceptible species,most cases occurred within 1-1.5 months after oestrus.In addition,the incidence rate of dogs and cats was the highest in the non reproductive history,indicating that the fertility can inhibit the occurrence of pyometra.In the cases with reproductive history,the proportion of non natural production(dystocia,caesarean section and induced labor)was the highest(66.67% of bitch,and 60% of cat).The results of ocular pathology showed that most of the cases had enlarged uterine horn,uneven uterine thickness,and the appearance of the uterus was pink or crimson.The distribution of capillaries in the outer wall of uterus is increased,and there is wave motion when touching the uterus;The contents of uterus with different properties can be cut along one side of the uterus,and the fluid in the uterus can be extracted.The collected contents are divided into mucus,pus and purulent blood according to the changes of observed result.Some endometrium has cystic hyperplasia,inflammatory focus and bleeding spots.Histopathological observation showed that there were inflammation a nd no inflammation in the uterine tissue.There are inflammatory uterine typical lesions can be seen glandular hyperplasia or atrophy,endometrial stromal glands and muscular layer can be seen varying degrees of inflammatory cell infiltration,mainly neutrophils and plasma cells,accompanied by bleeding and edema.Non-inflammatory endometrial mild atrophy of the glands with varying degrees of hyperplasia.Whether conservative treatment can be used in non-inflammatory cases is worthy of further investigation.The microbial species of the contents in the isolated uterus of dogs were detected.Staphylococcus aureus,Escherichia coli,Streptococcus,salmonella,Trueperella pyogenes,serratia marcescens,Klebsiella Trevisan,Enterococcus,Mycoplasma and Saccha romyce were detected by q PCR,The main microorganisms were Staphylococcus aureus(78.95%)and Escherichia coli(78.95%).Among them,Mycoplasma and Saccharomyce were detected for the first time in dog pyometra,and the role of them in pyometra needs furthe r study.Staphylococcus aureus,Escherichia coli,Streptococcus,salmonella,Trueperella pyogenes,serratia marcescens,Klebsiella Trevisan,Enterococcus were detected in the content of isolated feline uteri by q PCR.The main microorganism is Escherichia coli,accounting for 50%.Combined with the pathogenesis regularity,Histopathological observations and the types of microorganisms in the contents of uterus after the onset of pyometra,reasonable suggestions are provided for the diagnosis and treatment of pyometra of canine and feline in Harbin.
