Semen preservation is one important part of animal reproduction technology to improve the utilization rate of semen.Actually,in actual production,long-term storage of sperm will lead to changes in the biology and function of sperm cells.Simultaneously,there are few related studies about the effective preservation of acetylsalicylic acid(ASA),sodium salicylate(SS)and Y-27632 in sheep semen.This study randomly selected 10 Dubo rams as the test object to explore the effect of longterm storage of sheep sperm by adding different concentrations of ASA,SS,and Y-27632 at different temperatures.The aims of this test were to improve the sperm motility,acrosomal integrity,plasma membrane integrity,fertilization ability,and nuclear integrity of ram semen after long-term storage.The results of this research showed that the addition of ASA or SS at 20? can significantly improve the sperm motility after storage(P <0.05).And the motility of the 50 mM / L ASA group or the 20 mM / L SS group were significantly higher than the other groups(P <0.05).Also,the integrity of the acrosome and plasma membrane were significantly higher than control group(P <0.05);compared with the control group,there was basically no significant difference in the nuclear integrity rate,proving that the two composition did not damage the sperm nucleus under the experimental conditions.Under the storage condition of 4 ?,the results of ASA and SS were basically as the results of 20 ?.But effectively extended the preservation time.When stored for 13 d,the sperm motility under the treatment of 50 mM ASA and 20 mM SS reached 62.43 ± 3.32 and 50.00 ± 0.71(%),which was significantly higher than control group and the rest(P <0.05);Both can improve the sperm acrosome and plasma membrane integrity rate,and delay downward trend of the sperm acrosome and plasma membrane integrity rate.Y-27632,as a selective ROCK1(p160ROCK)inhibitor.In this test,20 mM Y-27632 significantly prolonged the storage time of sperm,and the motility reached 60.73 ± 3.97(%)when stored for 18 d,while the sperm motility of the control group was <10(%)(P <0.05);After storage for 13 d,the sperm acrosome integrity rate and plasma membrane integrity rate of the 20 mM treatment group were significantly higher than the control group(P <0.05);Compared with the control group,there was no significant difference in nuclear integrity rate,which proved that the Y-27632 did not damage the sperm nucleus under the experimental conditions.In summary,the appropriate amount of ASA and SS can significantly prolong the preservation time at 20 ? and 4 ?,and improve the sperm motility,acrosome and plasma membrane integrity,sperm motility and no damage nuclear.The storage effect of 20 mM Y-27632 at 4 ? is better than the former two.In addition,compared with Y-27632,the cost of ASA and SS in this experiment is extremely low,and can be applied in practice,and the preservation effect of Y-27632 on sperm is also worth studying in depth and exploring its mechanism of action.
