Pig artificial insemination technology is more and more widely used on production,semen preservation,as a key technology of artificial insemination,has been given more attention,but there are still many problems.Low temperature preservation is simple and economical,but it is still immature,and it is still in the screening phase of diluent at present,cryopreservation can go beyond all bounds,but the use of artificial insemination by frozen semen is estimated to less than 1% all over the world,While the preservation of semen technology is always restricting the effect of artificial insemination.In order to improve the utilization rate of artificial insemination and popularize the semen preservation technology,3 experiments has been studied in this paper,experiment 1 is taking Landrace semen as research object,with estradiol?testosterone?progesterone?ATP and BSA as cryoprotectants,added with different concentrations respectively in Tris yolk diluent?Modena dilution and Modena+sk dilution and low temperature storage,detecting and screening the best additive,the most effective concentration,and the best dilution;experiment 2 is taking Landrace semen as research object,adding different concentrations of glycerol,egg yolk,and different sugars into the Tris egg yolk dilution and cryopreservation,exploring the best concentration of glycerol and egg yolk,and the effect of different carbohydrates;experiment 3 is Landrace?Yorkshire?Duroc as research object,comparing the effect of artificial insemination between using frozen semen and fresh semen in pig production.The results were shown below:Experiment 1:optimum concentration and optimum additive: optimum concentration is 10ug/ml estradiol?20ug/ml testosterone?80ug/ml progesterone?0.025,0.05mg/ml of ATP?0.05,0.1mg/ml of BSA,progesterone is the optimum additive.Optimum dilution: the optimum dilution is Modena+sk dilution,followed by Tris dilution of the egg yolk,and finally the Modena diluent.Among them,the effect of Modena+sk+0.05mg/ml BSA combination on the sperm preservation at 5? was the best.Experiment 2: 3% glycerol had a significant effect on the improvement of semen quality,too much glycerol also showed its toxic effect,6%?12% glycerol did not improve the semen quality and even close to death.Contenting 20% of egg yolk has a significant improvement in the quality of semen,and 10%?30% egg yolk shows a poor effect on semen quality.In the dilute solution containing 3% glycerol?20% egg yolk,the effect of trehalose and fructose on the improvement is better than the other sugars,sucrose and lactose showed no significant difference effect of improvement,the effect of glucose on the semen quality is the worst.Experiment 3:There is still a gap between the effect of artificial insemination by using frozen semen and fresh semen,lower sperm vitality after thawing is one of the main reasons that lead to a poor reproductive performance,and not to be used in large-scale pig farms.Summary from the operation,the first frozen semen insemination time delay 3-6 hours is feasible,reproductive performance of Duroc sows the worst.Sow with minor genital tract disease,frozen semen is difficult to pregnant,but replaced with fresh semen can be conceived pregnant immediately.Conception is ideal in a suitable temperature.This study shows the effects of various additives in boar semen low temperature preservation,cryopreservation artificial insemination of using frozen semen.In the future,further explore the effect of other additives and combination of semen preservation,and further develop the operation rules in artificial insemination of frozen semen.
