Ovary,as an important reproductive system organ,plays its role by secreting reproductive steroid hormones and undertaking the process of follicular development.After sexual maturation,the follicles on the ovaries of mammals develop to ovulation stages,and the ovaries are divided into follicular phase and corpus luteum phase.In this study,the porcine ovary with similar age in different estrous cycles was studied.High throughput sequencing was carried out in the transcriptional group.The differential expression of porcine ovaries in the luteal and follicular phase was detected,and the differential expression genes(differentially expressed genes,DEG)were statistically analyzed,GO enrichment analysis and KEGG pathway were carried out.The purpose of this study is to explore the potential biological functions of target genes.The accuracy of RNA-seq was verified by real-time fluorescence quantitative analysis of 10 genes and sequencing results.The specific research content and results are as follows:(1)In the porcine ovarian transcriptional group in the luteal and follicular phase,4532 genes were expressed in two groups,85 genes were only expressed in the FP group,and the 851 genes were only expressed in the LP group.The differentially expressed genes were significantly enriched in 361 GO entries and 33 pathways,it mainly includes the biological pathways of carbon metabolism and fatty acid metabolism.(2)The analysis and screening of 17 candidate genes may play an important role in regulating follicular development,follicle atresia,ovulation,development and atrophy of the corpus luteum,steroid hormone secretion and other reproductive activities during the progesterone and follicular phase transition of the ovary.(3)10 genes(AREG,TM7SF2,SC4MOL,FDPS,HSD17B7,DHCR24,1DH2,CYP51,CTSL,SCD)were tested by qPCR test,and the results were similar to that results of RNA-seq sequencing.
