Effects Of Canine-derived Compound Probiotics On Intestinal Microflora,Digestive And Immune Function In Spleen Deficient Canines

The purpose of this study was to analyze the effects of canine-derived compound probiotics on the intestinal microflora,digestion and immune function in spleen deficient canines.Twenty-four Beagle canines with similar body weight,aged 1-year-old,were randomly divided into 4 groups,which were control group,spleen deficient group,spontaneous recovery group and probiotics treatment group,with six replicates in each group.The canines in control group were fed a basal diet;the canines in spleen deficient group,spontaneous recovery group and probiotics treatment group were fed the basal diet and were gavaged with 10 m L/(kg BW·d)senna decoction for 7days to establish spleen deficient model.On the day 7 of trial,the canines in spleen deficient group were dissected for collecting samples;the canines in control group and spontaneous recovery group were fed the basal diet for another 7 days,while the canines in probiotics treatment group were fed the basal diet and gavaged with 1×10~9CFU/d of canine-derived compound probiotics per canine for 7 days.The number of viable Bacillus bacteria in canine-derived compound probiotics was 6×10~7CFU/g,and the number of viable Lactobacillus bacteria was 1×10~8CFU/g.The experimental periods was 14 days.The results showed that:(1)Effects of canine-derived compound probiotics on the cecal microflora in spleen deficient canines.The ACE index,Chao1 index and Shannon index of the cecal microflora in the control group and the probiotics treatment group were higher than those in the spleen deficient group and the spontaneous recovery group,while the Simpson index was lower than that in the spleen deficient group and the spontaneous recovery group;Bacteroides at different classification levels such as phylum,class,order and family,and bacillus at class and genus levels,their relative abundances in the probiotics treatment group were significantly higher than those in the control group and the spleen deficient group(P<0.05),while the relative abundances of other microflora at different classification levels were not significantly different compared with the control group(P>0.05).(2)Effects of canine-derived compound probiotics on the digestive function in spleen deficient canines.On the 7th day,compared with the canines of control group,the contents of serum T-SOD,GSH-Px,D-Xylose and pancreatic protease were extremly significantly reduced(P<0.01),pancreatic amylase and lipase contents were significantly reduced(P<0.05),and MDA content was extremly significantly increased(P<0.01),duodenal and jejunum villi height and V/C values of spleen deficient canines were significantly reduced(P<0.05).As examined by transmission electron microscopy,compared with the canines of control group,the canines of spleen deficient group had sparse microvilli on the top of small intestinal mucosal epithelial cells,disrupted intercellular junction complexes,swelling of intracellular mitochondria and rough endoplasmic reticulum,and partial fusion degradation.On the 14th day,treatment with compound probiotics in the canines with spleen deficient symptom resulted in all the indexes were restored to the level of canines of control group(P>0.05).(3)Effect of canine-derived compound probiotics on immune function in spleendeficient canines.On the 7th day,compared with the canines of control group,the levels of serum Ig G,C4 and IFN-?in spleen deficient canines were extremly significantly reduced(P<0.01),Ig A,Ig M and C3 levers were significantly reduced(P<0.05);The positive expression level of s Ig A protein in the small intestine of spleen deficient canines was extremly significantly reduced by IHC method(P<0.01),the relative expression of Ig A m RNA in duodenum,jejunum and ileum of spleen deficient canines was extremly significantly reduced(P<0.01)by q PCR method.On the 14th day,there were no significant differences in the levels of multiple indicators between the probiotics treatment group and the control group(P>0.05).In summary,the canine-derived compound probiotics can restore the diversity of the cecum microbial of the spleen deficient canines,increase the proportion of beneficial bacteria such as Bacteroides,Bacillus and so on,and have a good regulation effect on the structure of the cecum flora of spleen deficient canines.The probiotics can restore the anti-oxidation,digestion and immune functions of canines with spleen deficiency to normal,and have a better therapeutic effect on canines with spleen deficiency.
