Dwarfing-closing planting,intensive cultivation due to premature fertility optimal production,is advantageous for the mechanized management advantages,has become the mainstream of current world apple cultivation mode.However,up to now,there is still a lot of uncertainty about the dwarfing mechanism of apple dwarfing rootstock,which makes the application of molecular breeding methods in dwarfing rootstock breeding process lack of sufficient effective means.This study biennial potted Fuji/SH40/MR(Malus robusta Rehd.),Fuji/M9/MR,Fuji/MR/MR 3 kinds of combination of rootstocks as the test materials,through the analysis of the transcripts sequencing in different combination of rootstocks of Fuji gene expression changes,to explore the M9,SH40 apple dwarf rootstock as interstocks on the regulatory mechanism of the scion.This paper aims to lay a foundation for the in-depth analysis of the dwarfing mechanism of two rootstocks and provide reference for the selection and breeding of dwarfing rootstocks.The main results are as follows:1.Transcriptome sequencing was carried out on the shoot-tips and phloem of red Fuji new shoot during the vigorous growth period.Results a total of 510 029 788 pairs of reads were obtained and the quality analysis results were reliable.2.There were 2108 differentially expressed genes in the transcriptome sequencing of the red Fuji interstock of M9 interstock and the red Fuji interstock of MR,among which 1005 genes were up-regulated and 1103 genes were down-regulated.The differentially expressed genes were involved in 111 metabolic pathways.Among the enriched KEGG pathways,the more significant and related metabolic pathways were brassinosteroid biosynthesis,carotenoid biosynthesis and carbon fixation in photosynthesis and nitrogen metabolism.SH40 interstocks of Fuji Fuji with MR interstocks transcriptome sequencing the differentially expressed genes in 3004,including 1798 genes expression,1206 gene expression.The differentially expressed genes were involved in 114 metabolic pathways,among which the enriched KEGG pathways were more significant and related to plant vegetative growth,including carbon fixation in photosynthesis,brassinosteroid biosynthesis,carotenoid biosynthesis,photosynthesis-antenna proteins.3.When M9 and SH40 were used as Fuji gene expression with MR interstocks compared with a total of 1060 differentially expressed genes,have been enriched to 77 metabolic pathways,the enrichment of differentially expressed genes in article 40 of the maximum number of metabolic pathways,strong correlation with plant growth and nutrition metabolic pathways have plant hormone signal transduction,two of the biosynthesis of terpenoids.carotenoid biosynthesis,carbon sequestration function of photosynthetic organisms and photosynthesis.4.QRT-PCR analysis was carried out on ABA1 SBPASE and RBCS1A in stem tip and phloem of red Fuji root in spring shoot growth period,spring shoot caping period and autumn shoot growth period of SH40 interstock,and it was found that the expression of ABA1,SBPASE and RBCS1A in scion could be improved when SH40 interstocks was used.5.The height,internode length and internode length of SH40 were significantly lower than those of MR interstock,but there were no significant differences in base stock diameter,scion diameter and new shoot length.6.The net photosynthetic rate of Red Fuji on SH40 was higher than that on MR.The results showed that the Red Fuji Pn of SH40 was higher than that of MR.7.The content of IAA in the red Fuji leaves of the interstock of MR in the spring shoot growth stage and the spring shoot capping stage was significantly higher than that of the red Fuji leaves of interstock of SH40.The content of ABA in the leaves of interstock of MR in the spring shoot growth stage,the interstock of the spring shoot capping stage and interstock of the autumn shoot was significantly higher than that of the middle anvil of MR in the spring shoot,the interstock of the spring shoot and the interstock of the autumn shoot.
