In Vitro Culture Of Meristem Tip And Rooting Technique Of Cut Chrysanthemum

There are many cut flower chrysanthemum varieties with high ornamental value.They are the main cut flower varieties in Ningxia and have high economic benefits and broad market prospects.At present,the production of cut chrysanthemum in China is introduced as the female parent,and the propagation of cuttings is used for cultivation,which is easy to cause virus accumulation in plants,resulting in quality degradation,and becomes the bottleneck of the production of cut chrysanthemum.In this study,six varieties of cut chrysanthemum were used as experimental materials to study the shoot tip culture and rooting technology in vitro,and to establish a complete system of virus-free seedling technology of cut chrysanthemum,which laid a foundation for ensuring the seedling quality,shortening the seedling period and improving the quality of cut chrysanthemum.The research results are as follows:1.Establishment of in vitro culture system of stem tip meristem of Cut ChrysanthemumStem tip peeling of two varieties ’Purple Dent’ and ’Amofei’ 3～4 mm is the best,and 4 varieties’Pink Dent’,’Spiritus Odoratus’,’Rebekai’ and’Mattis’ 4～5 mm works best.Callus and clump bud induction medium ’Purple Dent’ 6-BA 2.0 mg/L+NAA 0.3 mg/L+sucrose 25 g/L;’Pink Dent’ 6-BA 2.0 mg/L+NAA 0.2 mg/L+Sucrose 30 g/L;’Spiritus Odoratus’ 6-BA 2.0 mg/L+NAA 0.3 mg/L+sucrose 25 g/L;’Rebekah’ 6-BA 2.0 mg/L+NAA 0.3 mg/L+sucrose 25 g/L;’Amofei’ 6-BA 2.0 mg/L+ NAA 0.2 mg/L+Sucrose 35 g/L;’Mattis’ 6-BA 2.0 mg/L+NAA 0.1 mg/L+ Sucrose 30 g/L.Subculture proliferation medium ’Purple Dent’ 6-BA 0.5 mg/L+NAA 0.3 mg/L+sucrose 30 g/L;’Pink Dent’ 6-BA 0.5 mg/L+ NAA 0.3 mg/L+ sucrose 30 g/L;’Spiritus Odoratus’ 6-BA 0.5mg/L+NAA 0.15 mg/L+ sucrose 25 g/L;’Rebekah’ 6-BA 2.0 mg/L+NAA 0.15 mg/L+sucrose 45 g/L;’Amofei’6-BA 2.0 mg/L+NAA 0.15 mg/L+sucrose 45 g/L;’Mattis’ 6-BA 1.0 mg/L+ NAA 0.45 mg/L+sucrose 25 g/L.2.Establishment of rooting technology system of Cut Chrysanthemum in vitroThe optimum hormone level for rooting of the tissue culture seedlings of`Purple Dent’ and ’Pink Dent’ was 100 mg/L IBA,and the rooting rates were 91%and 96%,respectively.’Spiritus Odoratus’,’Amofei’ and ’Mattis’ were 100 mg/L ABT,and the rooting rate was 98%,98%and 95%,respectively.’Rebekah’ was 100 mg/L NAA,and the rooting rate was 92%.The best disinfectant for rooting matrix outside the bottles of ’Purple Dent’,’Pink Dent’ and ’Spiritus Odoratus’ was 0.1 g/L carbendazim,with the rooting rate of 98%,100%and 98%,and ’Rebekah’,’Amofei’ and ’Mattis’ were 0.1g/L potassium permanganate,with the rooting rate of 100%,98%and 98%.The cutting matrix of acupoint disc is peat:the ratio of perlite(3:2)was beneficial to the rooting outside the bottle of tissue culture seedling of chrysanthemum,and the rooting rate reaches more than 95%.3.Comparison of ornamental characters and quality performance of Virus-free Plantlets of Cut ChrysanthemumAfter 15 days of planting,the main growth characters of virus-free seedlings were higher than those of non virus-free seedlings;the flower characters of virus-free seedlings and non virus-free seedlings were basically the same in the four periods of budding stage,transparent stage,primary flowering stage and full flowering stage;the flower rate of A-level virus-free seedlings with cut flower quality reached over 94%,and that of B-level virus-free seedlings accounted for over 84%,and that of virus-free seedlings was better than that of non virus-free seedlings.
