Salix psammophila is one of the main tree species for desert control in northwestern China.It plays an important role in maintaining the stability of the sandy land ecosystem.It is an important element in urban construction and environmental greening projects.To promote the molecular directed breeding and study the regulation genetic mechanism,which is great significance for the development of the potential value of Salix psammophila.In this study,the genes of SpsNAC005,SpsNAC034,SpsNAC041,and SpsNAC042 cloned from Salix psammophila using homologous cloning technology were analyzed for bioinformatics,tissue-specific expression,and construction of over-expression vectors for target genes.The research results are as follows:?The CDS sequence length of SpsNAC005 gene is 927bp,encoding 309 amino acids,the CDS sequence length of SpsNAC034 gene is 1797bp,encoding 599 amino acids,the CDS sequence length of SpsNAC041 gene is 885bp,encoding 295 amino acids,the CDS of SpsNAC042 gene is 900bp and encodes 300 amino acids.They all have typical domains of NAC gene family and belong to NAC gene family.?RT-qPCR results showed that SpsNAC042 had the highest expression level in mature leaves and almost no expression in flowers.?In this study,we obtained 7 lines of SpsNAC005 overexpressed transgenic 84K poplar,5 lines of SpsNAC005 overexpressed transgenic Hebei poplar,5 lines of SpsNAC034 overexpressed transgenic Hebei poplar,4 lines of SpsNAC041 overexpressed transgenic 84K poplar,5 lines of SpsNAC041 overexpression transgenic Hebei poplar,9 lines of SpsNAC042 overexpressed transgenic 84K poplar lines,and 3 lines of SpsNAC005 overexpressed transgenic Hebei poplar.This study further enriched the molecular genetics information of the NAC gene family,laid the foundation for the functional study of the NAC gene family in Salix psammophila.To provided a theoretical basis for Salix psammophila.Provide a direction for the improvement of garden trees.
