Sequence Comparison Of Immune Related Gene Of Orf Virus From Different Hosts

The purpose of this study was to investigate the difference of ORFV immune related genes between virulent and attenuated strains and camels.This study,the virus genome was extracted from diseased sheep farms in Qingdao,from 90 generations of goat fibroblast cells and from bactrian camels in Alashan,the genome was named ORFV/QD/Y,ORFV/RD/Y,ORFV/QD/LT.Three pairs of specific primers were designed and synthesized according to the whole genomic sequence of sheep Aphtha published in Genebank,and the B2L,F1L and VIR gene fragments of ORFV/QD/Y,ORFV/RD/Y and ORFV/QD/LT were amplified respectively,the amplified DNA fragments were cloned into the PMD-19T vector,transformed into DH 5 ? Natural competence,and the recombinant plasmid was identified and sent to sequencing companies for sequencing,DNA Star software was used to splice the sequencing results and compare the nucleotide homology among the three genomes,the sequencing results were compared with the nucleotide sequences of thirteen ORFV genonies published on NCBI,and the phylogenetic tree and amino acid sequences were constructed.The reults showed that the nucleotide homology of B2L gene,F1L gene and V1R gene were 84.2%?96.6%,88.0%?98.3%and 77.4%?95.7%,respectively,the Nucleotide homology of B2L gene,F1L gene and VIR gene were 92.8%?99.2%,95.7%?100%and 77.0%?100%,respectively.The phylogenetic tree analysis of B2L Gene,F1L gene and VIR gene showed that ORFV/QD/Y was closely related to Chinese Fujian Goat Strain,ORFV/RD/Y was closely related to German sheep strain,and ORFV/QD/LT was a single branch,it was far away from other plants.The amino acid sequence of ORFV/QD/Y was compared with that of ORFV/RD/Y.The results showed that the amino acids of B2L and VIR were slightly different from ORFV/RD/Y,but the amino acids of F1L were significantly different,it may be related to the fact that the virus comes from the same species or has fewer generations.In order to explore the relationship between F11 and different species,further analysis of all 41 groups of F1L genes published in NCBI revealed that F1L genes showed significant differences among different species,the amino acid sequence of B2L and VIR genes of ORFV/QD/LT also showed significant difference,which further demonstrated the difference of ORFV infection among different species.The difference of these genes will lay a theoretical foundation for the future research of ORFV infection among strong and weak ORFV strains and different species.
