Spermophilus dauricus is a kind of animal of the genus daurian in the family sciuridae.It hibernates and prefers to live alone.It is an omnivorous animal with plant food as its main food.Spermophilus dauricus spends half of the year in hibernation,which has the typical characteristics of hibernation of mammals.There is a one-month difference between male and female individuals in the peak period of sleep,and they enter the mating season after hibernation.The duration of pregnancy is 25 days,and the lactation period can be up to one month.During hibernation,the external temperature of the Spermophilus dauricus can be as low as 2-8 degrees.The body functions of the Spermophilus dauricus are still active when they out of hibernation.After the low temperature in winter,the skin diseases caused by some parasites and bacteria are obviously improved.The effect of hibernation on the hair growth of Spermophilus dauricus has not been reported.In this paper,the PCR identification of the sex,the hair growth cycle and the full-length transcriptome were preliminarily observed,analyzed and studied,and the following results were obtained:First,the PCR amplification technology can use Smc8MD and Smc8MR1,Smc8MD and Smc8MR2,and Smc8MD and Smc8MR3 as the upstream and downstream primers to identify the sex of Spermophilus dauricus.Second,the coat growth cycle of Spermophilus dauricus1.The hair follicles of Spermophilus dauricus,like those of other mammals,undergo a growth cycle that includes phases of anagen prosperity,catagen,and telogen.2.The whole body of newborn Spermophilus dauricus was fleshy pink,and after 10 days,its back was obviously change into black.After 20 days,its dorsal surface was completely covered with hair,and it was obviously sandy yellow.After 30 days,the coat is visibly shaggy and the individual is similar to the adult.The dorsal hair of the Spermophilus dauricus is characteristic of the typical Agouti hair.Throughout the process,the hair follicles basically grow at the same time throughout the body.3,From the first year after hibernate to the second year before hibernate,the adult Spermophilus dauricus grows twice coat hair,experienced two hair cycles,the hair cycle is concentrated in the main point or a region,and then spread to the head and limbs.The whole process takes nearly 30 days from anagen to telogen,the telogen between two hair cycle,with the intermediate period ranging from 20 days to 30 days,and the second cycle time is the same as the first cycle time.Different from other species,the hair cycle of the back of the Spermophilus dauricus starts from the middle of the dorsal and spreads to the four sides until it reaches the extremities.There is no alternating situation,and there is no linear cycle field.The next hair cycle is the same.Third.Full-length transcriptome of the adult Spermophilus dauricus.Three generations of sequencing technology were used to extract RNA.The cDNA library was constructed by mixing the RNA in equal parts.The full-length transcriptome of the Spermophilus dauricus was 1997341369 bp.A total of 93621 transcripts were obtained after quality control.Gene annotation was applied to 20608 NR databases,31683 NT databases,19719 UniProt databases,and 14099 predicted ORF databases.2.In the GO database,76336 transcripts were annotated and divided into 65 branches.Among them,24342 transcripts were annotated for biological process,24843 transcripts for molecular function and 27151 transcripts for cell composition.21564 transcripts were annotated in the KEGG database.According to the metabolic pathway,it is divided into six categories.It is worth noting that the second of these six categories is the metabolic pathways related to human diseases,indicating that certain human diseases also occur in individuals of the Spermophilus dauricus.Compare UniGene with KOG database by default parameters of blast software,filter and annotate the optimal results.A total of 14687 transcripts were annotated,25 in total.A total of 4268 transcription factors and 63 families were predicted.Through the transcriptome analysis of the Spermophilus dauricus we found that there were RNA variable shear and a large number of redundant sequences in the full-length transcripts.At the same time,the analysis results show that the Spermophilus dauricus may have a unique metabolic pathway.
