In order to solve the doubts encountered in the application of probiotics in production and achieve the purpose of using probiotics and traditional Chinese medicine extracts reasonably.In this study,phenotypic characteristics and PCR technology were used to identify probiotics.Through antibacterial extrasomatic test,subculture test,bacterial growth curve and live count counting experiment,the culture of probiotic bacteria was screened and optimized to prepare probiotic powders and extracts of traditional Chinese medicine jointly created a compound probiotic product,and used mice to detect its safety,used metagenomic sequencing technology to monitor product quality,and used short-term animal experiments to initially explore the role of the product.The results were as follows:1.Two kinds of probiotics were selected from 7 strains of probiotics:Bacillus subtilis and Bacillus licheniformis.2.Optimized cultivation of Bacillus subtilis and Bacillus licheniformis:the basic nutrition was TSB,Bacillus subtilis added 1%peptone,3%glucose and0.3%Na H2PO4,Bacillus licheniformis added 1%peptone,1%glucose and 0.1%Na H2PO4.3.Preparation conditions of Bacillus subtilis and Bacillus licheniformis powder:96%corn starch,4%poloxamer,heat dried at 40?.4.Preparation of Shan Duo probiotics:Sophora subprostrate polysaccharide0.1%,mass ratio of Bacillus licheniformis powder and Bacillus subtilis powder was 1:1.5.Monitoring of Shan Duo probiotics:no toxic and side effects on mice;metagenomic sequencing showed no significant change in the proportion of probiotics before and after product preparation?P>0.05?.6.The role of Shan Duo probiotics:within 4 weeks,it could improve the SOD activity of Guangxi yellow chickens and the antibody titer of NDV?P<0.05?;the development data of immune organs in the Shan Duo probiotics'test group was higher than that in the blank control group,but the biostatistics results showed no significant difference?P>0.05?;no significant effect on the weight gain of Guangxi yellow chickens?P>0.05?;it could protect the intestinal flora and avoid the destruction of ampicillin on the intestinal flora,but the protective effect was not obvious?P>0.05?.Conclusion:1.The low temperature thermal drying method using 4%poloxamer as protective agent to prepare Bacillus licheniformis and Bacillus subtilis powder was feasible.2.Metagenome sequencing technology combined with live bacteria count counting method to jointly monitor the quality of probiotic products could achieve both qualitative and quantitative effects.3.Sophora subprostrate polysaccharide could be used in combination with Bacillus licheniformis and Bacillus subtilis,the composite product could improve the antioxidant capacity of Guangxi yellow chickens and enhance the humoral immune function within 4 weeks.It had no effect on the growth of Guangxi yellow chickens,and the recovery of the destroyed intestinal flora required long-term repeated experiments to verify.
