The experimental study of the effect of traditional Chinese medicine and probiotics on intestinal flora diversity and structure of rats in heat stress. It was designed to investigate the relationship between heat stress and the changes of intestinal, and then provide the theoretical basis of intervention mechanisms in heat stress for the traditional Chinese medicine and probiotics.80 rats were randomly divided into four groups, traditional Chinese medicine groups, probiotics group, heat stress group and the control group in normal temperature, and 20 rats in each group. The rats in traditional Chinese medicine groups were gavaged oral medicine according to 7mg/kg·BW rats weight, the probiotics group were gavaged the same volume as traditional Chinese medicine groups of 0.04% probiotics orally. In heat stress 3 hour, 1 day, 3 day and 7 day, determination of rat rectal temperature, concentration of White Blood cell, blood biochemical index, serum cortisol concentrations and the content of HSP70, observed the small intestinal mucosal morphology, gathered the cecal contents, by extracting the genomic DNA and construction of Library sequencing, Illumina miseq sequencing, obtained high quality sequencing data, the bioinformatics analysis, to know the effect of compound Chinese medicine and probiotics on animal the structure and the diversity of intestinal flora under heat stress.The results were as follow:In different experimental time, rats rectal temperature of traditional Chinese medicine groups, probiotics group and control group of were significantly lower than the heat stress group(P<0.05). After 3h, 1d and 7d, traditional Chinese medicine group, probiotics group and control group of the concentration of white blood cells in rats were significantly lower than the heat stress group(P<0.05). At 3 hours after the experiment, traditional Chinese medicine group and probiotics group ALB were significantly higher than the heat stress group(P<0.05). The ALT and AST in heat stress group were very significantly higher compared with the control group(P<0.01). Glucose content in traditional Chinese medicine group and probiotics group decreased markly significantly compared to the heat stress group and control group(P<0.01). At 7 days after the experiment, traditional Chinese medicine group, probiotics group and control group, their level of TP and ALB was drastic significantly higher than the heat stress group(P<0.01), and their ALT and AST levels were significantly below the heat stress group, compared with heat stress group, traditional Chinese medicine group, probiotics group and control group(P<0.01), the levels of blood glucose concentration were highly decreased significantly(P<0.01).The small intestinal villus epithelial cell degeneration in traditional Chinese medicine group, jejunum and ileum had no obvious pathological changes. Probiotics group and control group had no obvious histological changes in small intestine. In heat stress group, the intestinal villi of the duodenum were severely degenerated, and the epithelial cells of the jejunum intestinal villi were degenerated, some of the intestinal villi were broken, and the basal layer of the ileum was broken.Test in the 1day, 3day and 7day, traditional Chinese medicine group, probiotics group and heat stress group, the serum cortisol concentration were extremely significantly lower than that the heat stress group(P<0.01). At 3 hours and 3 day after the experiment, traditional Chinese medicine group, probiotics group and control group serum HSP70 concentration were highly significantly lower than the heat stress group(P<0.01).Traditional Chinese medicine group and probiotics group, intestinal bacteria group chao1 index and Shannon Wiener index of were significantly higher than that of heat stress group and the heat stress group chao1 index and PD whole tree index was significantly lower than that of normal blank group(P<0.05). PCA and PCo A analysis showed that traditional Chinese medicine group, probiotics group and heat stress group of intestinal bacteria colony began to appear difference, showing a tendency to separate. 7 days of experiment, control group and the other three groups scatter, heat stress group and the other three groups scatter and control group with heat stress group scatter completely separate, and traditional Chinese medicine group and probiotics group scatter basic overlapping.Under heat stress of gut microbes dominant bacterium had changed, Firmicutes and Bacteroides was relatively reduced(P>0.05), Lactobacillus of the genus was extremely significantly reduced(P<0.01), Clostridium increased, traditional Chinese medicine Group of Bacteroides and probiotics group of Lactobacillusa were significantly increased compare to heat stress group(P<0.05).Gene function of KEGG statistical analysis showed that heat stress in control group sugar metabolism, amino acid metabolism and membrane transport of gene expression of active, traditional Chinese medicine group, probiotics group and control group differences in the biological pathways was not significant, and traditional Chinese medicine group of biological gene activity on the path the lowest, probiotics biological pathways and room temperature close to the control group.
