| Through the separation and identification of soil collembola in the rhizosphere and non-rhizosphere of Acanthopanax senticosus,the analysis of soil physical and chemical properties and the extraction of root exudates,the analysis of the diversity change of soil collembola of the rhizosphere and non-rhizosphere in different months,the relationship between collembola and soil physical and chemical properties,and soil collembola and root exudates,to explicited the changing rule of diversity of the soil collembola in the root of Acanthopanax senticosus.Soil collembola of the rhizosphere and non-rhizosphere were collected by the soil drilling method,and the Tullgren funnel device separates the collembola and keep them in 75% alcohol.The isolated collembola were examined under a stereoscopic microscope to identify and count the collembola.The identification of the deep-colored collembola required decolorization,and the genus was identified according to the "Chinese Soil Animal Retrieval Guide" after the identification,the diversity indices of the soil collembola of rhizosphere and non-rhizosphere were both analyzed by DPS software,using one-way analysis of variance(One-way ANOVA)and LSD to compare the differences between different data groups.The statistical analysis of all data were analyzed and processed in SPSS22.0 statistical software,and the chart was made in Excel 2010.The rhizosphere soil was collected by the shaking method and analyzed by HPLC.Pearson correlation analysis and redundancy analysis were used to obtain the relationship between soil collembola and root exudates.Pearson correlation analysis and redundancy analysis were used to obtain the relationship between soil collembola and soil environmental factors.The results showed that 512 soil collembola were captured,belonging to 5 families and 5 genera.The root exudates of Acanthopanax senticosus were extracted and analyzed,to obtain three kinds of root secretions,it were salicylic acid,syringic acid,and gallic acid.The highest content of syringic acid,the total concentration was 57.13 mg/L.Salicylic acid content is second,the total concentration was 16.04 mg/L,and the concentration of gallic acid was the lowest,the total concentration was 4.80 mg/L.The contents of the three root exudates in the soil increasing trend with the month.There were significant differences in the species richness index,number of individuals,and evenness index of rhizosphere and non-rhizosphere soil collembola(P<0.05).In June,the functional trait value of soil collembola in rhizosphere was significantly lower than in the non-rhizosphere(P<0.05).Species richness and abundance of collembola has a very significant positive correlation with soil pH(P<0.01),and the dominance index has a significant negative correlation with gallic acid,syringic acid,and salicylic acid(P<0.05),Shannon-wiener diversity index was significantly positively correlated with soil pH(P<0.05),and significantly negatively correlated with syringic acid and salicylic acid(P<0.05).At the same time,we found that there was a significant difference of soil collembola between the rhizosphere and non-rhizosphere in June,and the rhizosphere effect of Acanthopanax senticosus on soil collembola was not significant in the July and August sampling.It showed that different sampling time and soil environmental factors affect the rhizosphere effect of Acanthopanax senticosus on soil collembola. |
