CircRNAs are involved in many important biological processes and play an important role as "molecular sponges" for miRNAs.The ovaries are important organs that produce eggs and secrete estrogen.There are few reports on the types,functions and regulatory mechanisms of circRNA in ovarian development.Therefore,this study examined and analyzed the expression of circRNA in ovarian tissue of estrus and estrus in dairy goats,and provided experimental basis for further exploring the regulation mechanism of circRNA in ovarian development of estrus cycle in dairy goats,and also to improve the reproductive rate of dairy goats.Provide new ideas and methods.In this study,the ovary of the estrus and interstitial cows was used as the research object.The whole library was prepared by extracting total RNA from the sample and processing it.The Illumina HiSeq was used for sequencing and analysis,bioinformatics methods,GO and KEGG pathway analysis,using RT-qPCR and other detection techniques,the circRNA in ovaries of milk goats during estrus and interestrus stage and its role in the estrus process of milk goats were studied,and the physiological mechanism of circRNA with different expression in ovaries during different development stages was predicted from the perspective of circRNA regulation.The main findings are as follows:1.Screening and validation of circRNAs in ovary of dairy goats during estrusThe results showed that there were 22,333 candidate circRNAs in ovarian tissue of estrus and estrus in frogs,which could be divided into six categories.There were 772 circRNA differences in ovary relative estrus in estrus in dairy goats.Expression,268 up-regulated,504 down-regulated;in addition to the Y chromosome in ovary tissue of dairy goats,circRNAs are widely distributed on all other chromosomes,and most circRNAs are approximately 500 nt in length.2.Predictive analysis of related circRNAs in ovary of estrus in dairy goats(1)GO analysis showed that a total of 86 groups were significantly enriched in cellular components.In the molecular function GO analysis,circRNA was significantly concentrated in 111 classifications.In the bioprocess classification,it was clustered into 371 groups.(2)KEGG results showed that 105 pathways were significantly enriched,including adhesion junctions,ubiquitin-mediated proteolysis,and endocytosis.This study investigated the expression levels of circRNA in ovarian tissue of dairy goats at different developmental stages.CircRNAs may be involved in ovarian development-related signaling pathways,which may be related to the estrus of dairy goats.It opens up a new field for the function of circRNA in ovarian development and provides data support for the specific biological functions and molecular mechanisms of circRNA involved in ovarian development and estrus in dairy goats.
