| Pigs are very similar to humans in terms of physiology and anatomy,so they have become a more commonly used large-scale experimental animal in medicine and veterinary Medicine.Inbred animals have high homozygosity,consistent phenotype and clear genetic background,which have shown strong advantages in the application of scientific research,so they have become the main direction and hot topic of experimental animal breeding.Currently,most of the inbred animals that have been successfully cultivated are mainly mice and rats at home and abroad,while there are relatively few lines of inbred pigs.Domestic researchers have bred inbred pigs such as Wuzhishan miniature pigs,Bama miniature pigs and Banna Mini-pig,which were all inbred with local pig breeds which live in low altitude area.However,there are no inbred pigs in plateau area internationally.Hezuo pigs is a kind of plateau’s original minipigs which live in Gannan Tibetan autonomous prefecture,carrying out standardized breeding of experimental animals on Hezuo pigs can fill the defects and deficiencies in plateau’s inbred minipigs.In this study,blood routine,blood biochemistry and genetic markers of microsatellite sites were detected in Hezuo pigs of a dozen generations of inbreeding,and compared with the same indexes of original HeZuo pigs.The aim of our study is to explore the physical and genetic characteristics of inbred Hezuo pigs and provide theoretical basis and reference for the establishment of Hezuo pigs inbred lines.First,we analyzed the physiological performance of inbred Hezuo pigs and original HeZuo pigs.The test of 12 blood routine and 16 blood biochemistry indicators were used to analyze the biological differences including body’s health status,metabolic level and immune function between inbred Hezuo pigs and original HeZuo pigs.Results showed that after decades of inbreeding,Hezuo pigs still maintain their original physiological and biochemical characteristics.Among them,the characteristics peculiar to plateau animals are more prominent,which not only reflects the better offsite adaptability and growth potential of Hezuo pigs,but also shows the uniqueness of Hezuo pigs as experimental animals.Secondly,we analyzed the genetic characteristics of inbred Hezuo pigs and original HeZuo pigs.After synthesizing 10 pairs of primers,we use microsatellite DNA technology to detect the number of alleles,effective allele number,heterozygosity,hexp and Polymorphism information content in microsatellite sites.Results shown that the number of alleles in inbred Hezuo pigs is significantly less than that in original HeZuo pigs,and inbred Hezuo pigs are more homozygous.The number of moderate and low polymorphic loci in original HeZuo pigs are also much higher than inbred Hezuo pigs,which shown that inbred Hezuo pigs are expected to become a new inbred minipig strain after continued breeding for several generations.This research shows that inbred Hezuo pigs still retain the unique physiological and biochemical characteristics of original HeZuo pigs,and the number of alleles-have decreased,while genetic homogeneity increased.Therefore,inbred Hezuo pigs are expected to be bred into a new breed experimental animal and used in biological science. |
