| In this study,the morphology and molecular markers of the Dermacentor nuttalli and Dermacentor silvarumv,and the structure of the midgut contents were studied.The results are as follows:The surface morphology and structure of the Dermacentor nuttalli and the Dermacentor silvarumvă were observed and compared by stereo microscope,and then the molecular markers of the two were compared based on the COX1,ITS2 and 16 S rDNA gene sequences.The results showed that the morphology and structure of the two alfalfa were different,mainly in the three aspects of body type,tannin and margin.However,the sequences of COX1,ITS2 and 16 S rDNA were very similar,and the similarity was 99.76% and 99.75%.98.29%.The PCR-DGGE method and high-throughput method were used to analyze the structure of the contents of the midgut contents of the Dermacentor nuttalli and Dermacentor silvarumv.The results of PCR-DGGE analysis were: Rickettsia barbariae,Coxiellaceae bacterium,Escherichia coli,Anaplasma phagocytophilum are two kinds of midgut contents.There are a total of dominant bacteria.The molecular results of the high-throughput method are: Proteobacteria is the most abundant gate;Rickettsiales is the most abundant;Anaplasma is in the engorgement Dermacentor nuttalli,and the Rickettsia genus The engorgement Dermacentor silvarumv,the half engorgement Dermacentor silvarumv and the half engorgement Dermacentor nuttalli have the highest abundance. |
