Using maize pollen to induce wheat double haploid can shorten the breeding period,speed up the breeding process and improve the breeding efficiency.In recent years,many scholars have conducted many studies on the use of maize pollen to induce wheat haploid,but in this technical system,the induction of wheat haploid embryos,the conditions of wheat haploid embryo germination and seedlings and the doubling of chromosomes and the treatment concentration of the plant of wheat haploid and other issues still needs further exploration and improvement.In this study,wheat hybrid F1 generation and dwarf wheat were used as test materials,using maize variety NK 18 as pollen donor,induction of wheat haploid embryos and chromosome doubling of wheat haploid plants,while obtaining wheat DH line?Double Haploid?,provide technical reference and method guidance for wheat double haploid.The results of this study are as follows:?1?In the florescence adjustment experiment,the flowering period of maize transplanted in greenhouse on January 14 and 21,2019 could meet with that of wheat planted in the field on October 20,2018.In this study,when the difference in sowing time was 92-99 days,the florescence basically met.?2?The cultivation method has a significant effect on the embryo yield of pollination ears.The in vitro culture rate of pollination ears?24%?is about twice that of natural field growth conditions?11.87%?.?3?Temperature has no significant effect on the germination rate of wheat haploid immature embryos.At 4?,the average germination rate of immature embryos of 10 hybrid combinations was 32.52%;at 25?,the average germination rate of immature embryos was33.02%;statistical analysis found that genotype had a significant effect on the germination rate of wheat haploid immature embryo.Among the 10 hybrid combinations,the germination rate of material 19F1-35?4?,25%;25?,20.83%?was lower than others,and the germination rate of material 19F1-59?4?,37.50%;25?,43.18%?was higher than others.?4?Wheat haploid immature embryos produced by induction of dwarf defeat wheat were inoculated into 1/2MS medium and 2/3B5 medium respectively,analysis of variance shows,there was no significant difference in the germination rate and seedling rate of immature embryos on these two mediums.The germination rate of haploid immature embryos on 1/2MS medium is 41.67%,and the germination rate on 2/3B5 medium is35.42%,the former is 1.18 times that of the latter;The seedling rate on the medium is35.42%,and the seedling rate on the 2/3B5 medium is 25%,the former is 1.3 times that of the latter.?5?Studies on chromosome doubling of wheat haploid plants found that colchicine with a concentration of 8 g/L added to the surface of the medium for 24 h had the best doubling effect,and the doubling efficiency reached 14.7%.
