| Zanthoxylum planispinum var.daintinesis,an ecological management plant in the karst stone desert area of Guizhou,has become an important characteristic agricultural product depended on its important economic value.Being famous for its strong numbness,the content of pungent constituents of the plant is directly related to the quality of the pericarp products.Due to the difficult separation and instability of the pungent standard substances,the quantitative evaluation of pungent in zanthoxylum samples is very difficult,resulting in not only quality evaluation of the pungent products,but also the research on the relationship between quality and factors associated with its growing environment.In this thesis,the quantitative analysis method of pungent constituents in Z.planispinum var.daintinesis was studied,and the quality evaluation and the correlation between the pungent components and the soil environmental factors were preliminarily studied.Some research results were obtained:1)A method for the rapid separation of hydroxyl-α-sanshol and hydroxyl-β-sanshol in tingtan prickly ash was proposed by the combination of High-speed Countercurrent Chromatography and high performance liquid chromatography.2)Based on "QAMS"(quantitative analysis of multi-components with a single-marker),the content of 6 pungent componets in the Z.planispinum var.daintinesis were simultaneously determined with hydroxyl-β-sanshol as internal standard,and the results showed that the purity of standards affected the results of the sharply.At the same time,it was shown that in the prickly ash pericaps,the principal components,determined as hydroxyl-α-sanshol and hydroxyl-β-sanshol,can represent the basic pungent components.3)Hydroxyl-α-sanshol,hydroxyl-β-sanshol and hydroxyl-ε-sanshol,the three isomers of the pungent component in the Z.planispinum var.daintinesis wer considered as the evaluation index for the pungent content.Using hydroxyl-β-sanshol as internal standard,a simple analysis protocol was proposed based on "QAMS",with the necessary of the expensive standards,hydroxyl-β-sanshol;4)A method for the determination of numb-taste substances in Z.planispinum var.daintinesis based on "QAMS" was set up,using complex standards directly without complete separation of the isomers of Hydroxyl-α-sanshol,hydroxyl-β-sanshol.This provides a solution to the problems of difficult preparation,preservation and high cost of standard substances in the determination of numb-taste substances.5)Based on the components of numb-taste,HPLC fingerprint analysis method was established,and the identification model was provided for the qualitative evaluation of quality of the products from Z.planispinum var.daintinesis,according to the component characteristics of numb-taste components;6)By using water as solvent,a fast and green subcritical water extraction method for the pungent components from Z.planispinum var.daintinesis was proposed,which provides ideas for the deep development and utilization of numbness components.7)The correlation between the pungent component and the soil nutrients was discussed,and it provide a way to study the correlation of pungent component with the environment factors,although the results could not effectively reflect the correlation between them. |
