Hydrogen Pyrophosphorylase(H~+-PPase)is a type of H~+ transporter,it's different from H~+-ATPase,which has a significant on the physiological and biochemical processes in plants.As a proton pump,H~+-PPase plays an important role in plant stress resistance.The function of Ed HPPase1 gene expression in model crops has been verified,however,the physiological and regulatory mechanisms of crops have rarely been shown.In order to better explore the function of Ed HPPase1 in crops,our study will be used to verify in two ways under abiotic stress by hydroponic experiments and field experiments,respectively.Bioinformatics analysis shows that the open reading frame of the Elymus pylori pyrophosphorylase(Ed HPPase1)gene has a total of 2310 bp and encodes 770 amino acids,and Ed HPPase1 has 15 transmembrane spiral structures.The amino acid sequence alignment structure shows that it contains a complete conserved domain.The comparison of protein homology was found to be similar to known Leymus chinensis the homology of the inorganic pyrophosphorylase protein of barley is as high as 98%.Physiological indicators showed that the fresh weight content and relative water content of transgenic wheat were significantly higher than wild type,and the water loss rate was slower than wild type by alkaline stress;The root scan results showed that the expression of the alkaline grass inorganic pyrophosphorylase(Ed HPPase1)gene significantly increased some root indicators such as the projected area,surface area and volume of transgenic wheat roots compared with wild type under the hydroponic experiments.However,the transgenic wheat lines had no difference in plant height and fresh weight content from the wild type,and the root length and root fresh weight content were lower than the wild type lines under the normal treatment;The biochemical index measurement results showed that the content of soluble sugar,proline and soluble protein in the transgenic wheat increased significantly compared with the wild type,while the content of malondialdehyde and relative conductivity decreased compared with the wild type;The antioxidant enzyme indicators showed that the contents of SOD,CAT,POD and superoxide anion radicals in transgenic wheat were significantly increased compared with wild type under alkaline stress.The field experiment's results showed that the content of total nitrogen and total phosphorus in the straw and grain of transgenic wheat was significantly higher than wild-type line,while the content of straw and grain of transgenic wheat and the change of yield as same as Low-P treatment,and the yield increases slightly lower than Low-P treatment.In conclusion,the expression of Ed HPPase1 gene increased the nutrient content of wheat at harvest time under the low phosphorus treatment.
