Psastachys Huashanica,a specific wild relative of wheat in China,has been reported to exhibit a variety of disease-resistant and stress-resistant traits.H-6-11-1 is a F8 generation derived progeny obtained by hybridization of common wheat 7182×Psastachys Huashanica and continuous self-cross.And it had excellent resistance to sharp eyespot.In this study,we intend to investigate and analyze data related to disease resistance and agronomic traits of H-6-11-1.In addition,molecular cytogenetic analysis,SSR marker analysis and other methods were used to determine the genetic composition of H-6-11-1,so as to determine whether there were imported exogenous chromosome fragments.And we construct the H-6-11-1 x Yang mai158 F2 segregation population to analyze the optimal inheritance model of resistance.Moreover,the resistant bulk and susceptible bulk were established by using Bulked sample analysis?BSA?.The DNA of the corresponding material was extracted for 660K array scanning,and combining the SSR markers screening.Through these above methods,we preliminarily localize the chromosomal locations of the genes related to sharp eyespot resistance.And it can be used as the theoretical basis and material support for the discovery and functional verification of candidate genes for sharp eyespot resistance.The results in this study are as follows:1.The results of disease resistance identification showed that H-6-11-1 had good comprehensive resistance.It was moderately resistant?MR?to sharp eyespot,resistance?R?to FHB,high resistance?R?to powdery mildew,and near immunity to stripe rust.In addition,7182 and Yangmai 158 were susceptibility?S?to sharp eyespot.So it was concluded that the resistance of the wheat-Psastachys Huashanica introgression line H-6-11-1 was derived from its exogenous parent Psastachys Huashanica.Through the statistical analysis of the agronomic traits of H-6-11-1,We found that it had a smaller plant height and a higher thousand-seed weight,and met the standard of high-quality strong gluten wheat.So we think the comprehensive agronomic traits of H-6-11-1 were good.2.Cytogenetic analysis of H-6-11-1 showed that the total number of chromosomes of H-6-11-1 root tip cells was 42,and no obvious yellow-green exogenous chromosome or exogenous chromosome fragment signals were found in in-situ hybridization.The possible reason is that too small exogenous fragments were imported.Combined with SSR markers,660K array scanning and disease resistance phenotypic analysis,it was speculated that H-6-11-1 was the progeny material of the wheat-Psastachys Huashanica introgression line.3.When we analyzed the genetic model of resistence to H-6-11-1,we found that the sharp eyespot resistance was controlled by two major genes.And we chose 2MG-ADI?two major genes-additive-dominant-epistasis?as the optimal inheritance model of resistance.According to the results of scanning 660K array,we speculated that the location of the resistance genes related to the disease was on chromosome 3BL and 5B.We used SSR markers distributed on 3BL and 5B chromosomes to screen for polymorphism.The result showed that in the three pairs of SSR markers?Xwmc687?Xwmc326?Xgwm547?on the3BL chromosome had different bands.We hypothesized that a new resistance-related QTL may exist in this segment.
