Abstract:Both American ginseng (Panax quinquefolium L.) and ginseng(Panax ginseng C.A.Mey.) are Araliaceae plants and rare traditional Chinese medicine. Their dried roots or rhizome are common medicinal parts. For the effective protection, development and utilization of medicinal herbs resources in recent years, the investigation emphasis were more focused on stems and leaves of these plants, material base, pharmacological activities and mechanisms of action, etc about American ginseng and ginseng are widely studied.it is common accepted that ginsenosides composition is the main pharmacological active ingredients. UPLC-ELSD fingerprints and content determination methods of seven ginsenosides in roots, stems and leaves of American ginseng and ginseng herbs will be established in our investigatin.Methods:we investigated solvents, extraction methods, extraction time, and other pocess parameters to establish a optimum extraction method. Then the optimum chromatographic conditions were fixed as follows:Agilent ZORBAX SB-C18column (4.6x50mm,1.8μm), a column temperature of25℃, and a gradient elution of acetonitrile (A)-water (B) as mobile phase at the flow rate of1.0mL/min. Parameters of evaporative light scattering detector were fixed on a drift tube temperature of40"C at gas pressure of0.32MPa and a gain value of7. In the methodological study, the results of resolution, precision, repeatability and stability tests complied with the provisions of TCM fingerprint and content determination.Result:(1)8,6,10common peaks were respectively identified with the peak of ginsenoside Rb1, Re, Rb3as reference in American ginseng roots, stems and leaves, which accounted for98.20%,96.39%,95.71%percent of total peak area; Fingerprints of American ginseng from Jilin similarity were more than0.90; American ginseng roots can be divided into two classes in accordance with the growth period:and American ginseng leaves can be divided into two classes in accordance with the localities.(2)8,3,10common peaks were respectively identified with the peak of ginsenoside Rb1, Re, Re as reference in ginseng roots, stems and leaves, which accounted for94.93,96.06%,97.78%percent of total peak area; Fingerprints of ginseng from Jilin similarity were more than0.90;but fingerprints of the biennial ginseng roots from Jilin similarity were0.8or so; ginseng roots can be divided into three classes in accordance with the growth period; and ginseng stems can be divided into two classes in accordance with the localities;and ginseng leaves can be divided into two classes in accordance with the localities.(3) with ginsenoside Rg1,Re,Rb1,Rb2,Rb3,Rd and pseuoginsenoside F11as index ingredients,the differences of ginsenoside contents in roots, stems and leavesof American ginseng and ginseng were compared on the basis of content determination. Research results showed that there are significant differences in ginsenoside contents of various herbs. Leafy medicines are rich in ginsenoside Rg1、Re; pseuoginsenoside F11is peculiar to the American ginseng ingredients, the highest level in American ginseng leaf; ginsenoside Rb1is distributed mainly in roots; leafy medicines and ginseng root are rich in ginsenoside Rb2; ginsenoside Rb3is distributed mainly in American ginseng leaf; ginsenoside Rd is widely distributed.Conclusion:UPLC-ELSD fingerprints and content determination methods of seven ginsenosides in roots, stems and leaves of American ginseng and ginseng herbs were established in our investigatin. The study provides a new hyphenated technique for ginsenosides. It will lay the foundation for the provenance identification, comprehensive quality control and evaluation, and in-depth research and development of American ginseng and ginseng.
