Objective Through experiments in the body,to study the function of the different sets of Radix Ginseng Rubra preparations for estrogen receptor positive breast cancer cells MCF-7 cell apoptosis and cycle derection,and the effect of estradiol level and staining pathology of the tumor tissue,to explore the impact of Radix Ginseng Rubra plant estrogen effects on human breast cancer MCF-7 a tumor-burdened nude mice subcutaneously transplanted tumor proliferation and apoptosis,in order to provide more theoretical basis for Chinese medicine clinical use of red ginseng,improve treatment evidence and treatment effect.Method Copy tumor-burdened models:Taking the logarithmic phase of MCF-7 cells,prepare with saline to(1-2)×108 cells/ml concentration of the cell suspension,implant them under the botton of right breast,inject 0.2ml cell suspension per mouse,make the tumor-burdened nude mice models.When the tumor grows to about 100mm3 randomized to the control group,estrogen control group and tamoxifen control group which are as two positive control groups,and the high,medium and low doses of Radix Ginseng Rubra preparation group.14 days later,after the program administered by a predetermined design,remove the nude mouse mammary tumor.Observe the general situation,measure the weight of nude mice regularly.Measure the tumor inhibition rate and the development of the growth curve.Use the enzyme-linked immunosorbent assay(ELISA)to detect serum estradiol levels.Cell apoptosis and cell cycle derection by Flow cytometry.Obeserve the Pathological morphology of the tumor issue.Reaults ?Through observation of tumors in general and tumor inhibition rate and growth curve,high doses of Radix Ginseng Rubra can inhibit the proliferation of breast cancer MCF-7 cells ? Detecteing by ELISA note,medium and high doses of Radix Ginseng Rubra lower the estrogen levels.And more obvious in medium dose group.Low doses of Radix Ginseng Rubra preparations elevate estradiol levels.(?)By flow cytometric detection of apoptosis results,various concentrations of Radix Ginseng Rubra can inhibit tumor,but only the high dose of Radix Ginseng Rubra can induce apoptosis.?The high doses of Radix Ginseng Rubra group can block G0/G1 phase significantly.?The pathological biopsy of tumor shows various concentrations of Radix Ginseng Rubra are related to tumor necrosis,where high concentrations are most obvious,the area is taken up more than 30%.Conclusion Radix Ginseng Rubra in high,medium and low doses can inhibit the proliferation of MCF-7 cells,high doses of red ginseng in MCF-7 block the G0/G1 phase and induce apoptosis.In the high-dose red ginseng has lowered estrogen levels.And more obvious in dose dose group.Low doses of red ginseng preparations elevated estradiol levels.And more obvious in medium dose group.Low doses of red ginseng preparations elevate estradiol levels.
