Structure Characteristics And Immunomodulatory Activities Of Polysaccharides From Radix Ginseng Rubra

Objective: In this experiment,two different polysaccharides were purified from Radix Ginseng Rubra,the structure of the two purified polysaccharide fragments was characterized and analyzed,and the immunomodulatory ability and anti-cancer ability of the two purified polysaccharide fragments in Radix Ginseng Rubra were explored.Methods: Radix Ginseng Rubra polysaccharides(RGRP)crude polysaccharides were extracted from red ginseng by hot water extraction,ethanol precipitation and Sevag deproteinization,and further purified by DEAE ion exchange chromatography column and S-300 gel chromatography column to obtain red ginseng polysaccharides(RGRP-1a and RGRP-1a).RGRP-1b).Monosaccharide composition,Methylation and GC-MS analysis,FESEM,AFM,FT-IR and NMR spectra were used to establish the structure of RGRPs.The aptitude of immune regulation and anti-cancer ability were explored by using different methods.Results: RGRP-1a and RGRP-1b have different molecular weight of21.3 KDa and 10.2 KDa.RGRP-1a consisted of glucose.However,RGRP-1b mainly made up of glucose,galactose and arabinose.RGRP-1a was predicted to be linear α-(1→4)-glucans with α-Glcp-(1→ attached to C-6 of(1→4)-glucosyl residues.The main chain structure of RGRP-1b was 1,4-α-Glcp,with a 1,4,6-α-Glcp branch unit.The branch is linked to O-4 a.The O-4 branch was composed of 1)-β-Galp-(4→1)-α-Araf-(5→α-Araf or 1)-β-Galp-(6→α-Glcp.Change of NO and cytotoxicity assay showed that macrophages might be activated by RGRP-1b which showed more powerful stimulating role on cell proliferation.The expression of IL-6,IL-12 and TNF-α was observed to be upregulated after the treatment with RGRP-1b.Conclusion: The findings indicate that RGRP-1b from radix ginseng rubra is of potential value of immune regulation.
