Objective:The proliferation and differentiation of neural stem cells in rat brain tissue were detected by immunofluorescence,and the different time points(6 hours,12 hours,24 hours,and 48 hours)of electroacupuncture at the time of neural stem cell transplantation for cerebral hemorrhage in rats were studied.Effect of differentiation of neural stem cells in rat brain tissue.To explore the best intervention time of electroacupuncture in the treatment of cerebral hemorrhage,and to explore the mechanism of acupuncture combined with neural stem cell transplantation in the treatment of neural repair and regeneration after intracerebral hemorrhage.Methods:160 Sprague-Dawley rats were randomly divided into transplantation group,sham operation group,model group,and electroacupuncture group.The intervention time of electroacupuncture was divided into 6 hours subgroup,12 hours subgroup,24 hours subgroup,and 48 hours.In the subgroup,the treatment time was two weeks.The rat ICH model was established using autologous blood injection and Berderson score was used to assess the behavioral changes.Rat hippocampal neural stem cells were isolated and cultured and identified by Nestin immunofluorescence with Brdu labeling;Brdu-labeled neural stem cells were transplanted into cerebral hemorrhage region of rats and sacrificed after 2 weeks.All brains were fixed,frozen sections were made,and immunofluorescence staining was performed.NeuN,GFAP positivecell count.Results:1.After the model of cerebral hemorrhage was awake,the difference between the model group,the electroacupuncture group,and the sham group was significant(P<0.05).The model group,electroacupuncture group,and transplantation group were directly compared with each other without significant difference(P>0.05).2.Change and comparison of rat GFAP positive cell number: After immunofluorescence staining of rat brain tissue,GFAP positive cells were seen in sham group,model group,electroacupuncture group and transplantation group,and EA group was transplanted at the same time point.The number of GFAP-positive cells was higher than the other three groups.There were differences between the groups,and the difference was statistically significant(P<0.05).The number of GFAP positive cells at each time point in the transplanted electroacupuncture group was compared.The number of subgroups at the 6th hour was the highest,and there was no significant difference compared with the 3 subgroups at 12 h,24h,and 4h(P>0.05).3.Changes and comparison of rat NeuN positive cell number: After immunofluorescence staining of rat brain,NeuN-positive cells were seen in the sham-operated group,the model group,the transplanted electroacupuncture group and the transplantation group,and the electroacupuncture group was transplanted at the same time point.The number of NeuN-positive cells was higher than the other three groups.There were differences between the groups,and the difference was statistically significant(P<0.05).The number of NeuN-positive cells at each time point in the EA group was compared.The number of subgroups in the 48-hour subgroup was the highest,and there was no significant difference between the subgroups at 6h,12 h,and 24h(P>0.05).Conclusions:1.Observe the degree of neurological deficit after intracerebral hemorrhage in rats in each group and find that the electroacupuncture group can significantly improve the neurobehavioral performance after intracerebral hemorrhage in rats.2.The highest expression of 2.6hGFAP was observed,and the expression ofNeuN was the highest at 48 h.Electro-acupuncture had a repairing effect on brain tissue injury after intracerebral hemorrhage in rats.The mechanism may be to promote the differentiation of NSCs into neurons and glial cells.
