Objective Through the detection of the quantity of Nestin expression in brain tissue in model rats with ICH(Intracerebral hemorrhage,ICH),we study the treatment of ICH and to explore the mechanism of Electro-acupuncture and neural stem cells transplantation in the treatment of ICH.Methods The 160 rats were randomly divided into four groups: model group,sham operation group,transplantation group and electro-acupuncture group.Each group was then divided into four groups,namely 6h,12 h,24h and 48 h,respectively,according to the time of intervention of the electric needle.Caudate nucleus of autologous blood injection method is used to replicate hemorrhage rats model,according to bederson score method to screen out the successful model,after the success of the building on the third day in the caudate nucleus position transplanted neural stem cells,the curative group in transplant success after 6 h,12 h,24 h,48 h electric acupuncture intervention,four time points after two weeks of electric acupuncture intervention by rt-pcr method the expression of nestin,testing rats had after the amplification results using relative quantitative method 2-??Ct value to compare the relative gene expression.Results1.After 2h of cerebral hemorrhage in rats,the model group,transplantation group and electro-acupuncture group showed different levels of nerve damage,which was statistically significant compared with the sham group(p < 0.05).Electric-acupuncture group after neural stem cell transplantation,electric acupuncture intervention at various time points,the rat nerve defect signs were improved,and compared with before treatment,the difference is statistically significant(p < 0.05),compared with model group,there was no statistically significant difference(p > 0.05).2.Comparison of gene expression levels in each group after RT-PCR amplification:2.1 For the same time point nestin protein expression of four subgroups compared respectively,nestin protein expression of electro-acupuncture group were higher than that of the rest three groups,and 6h electro-acupuncture group compared with the same point in time transplantation group,the difference statistically significant(p < 0.05).12 h,24h,48 h electro-acupuncture group compared with the corresponding transplantation group,the expression of the nestin protein was increased,but not statistically significant(p > 0.05).2.2 in the group and the acupuncture group each time point nestin protein expression,6 h subgroup nestin protein expression quantity highest,with 24 h and 48 h subgroups compared statistically significant(p < 0.05),compared with 12 h subgroups,no statistical significance(p > 0.05).There was no statistical significance(p >0.05).Conclusions Compared with transplantation group,the curative group of each subgroup nestin protein expression quantity increased significantly,showed that after cerebral hemorrhage curative intervention for the repair of the nervous system has obvious effect,at the same time in every time subgroup of cupping treatment,the curative group nestin protein expression of the overall trend for electric acupuncture intervention in the earlier time,the higher the nestin protein expression quantity.The most obvious increase was in the 6h subgroup,which indicated that after the transplantation of neural stem cells 6h began to be treated with electro-acupuncture,the effect was most obvious.
