From BDNF,NGF And Nestin Angle To Study The Protection And Repair Mechanism Of Electro Acupuncture On The Neural Damage Of Brain Ischemia

Objective : From BDNF , NGF and Nestin angle to study the protection and repair mechanism of electro acupuncture (EA) on the neural damage of middle cerebral artery occlusion (MCAO) rat. And try to study if acupuncture serum can influence mesenchymal stem cells differentiate into neuron-like cells.Method : (1) Zea-Longa method was adopted to MCAO models. (2) To observe the improving of neural defective symptoms estimated by neural behaviors. (3) To observe the dynamic changes of pathology with Hematoxylin-Eosin staining. (4) Enzyme linked immunosorbent assay (ELISA) was adopted to examine brain-derived nerotrophic factor (BDNF) of ischemic cortex , and to study the changing of BDNF after the EA treatment . (5) Immunohistoche micalassay (SABC) was adopted to detect the expression of nerve growth factor and nestin of the ischemic cortex and the changing after the EA treatment. To analyze the EA protection to the neural damaging after the ischemia. (6) To study if the EA serum and can influence mensenchymal stem cells (MSC) differentiate into neuron-like cells or not, to do some basal experiment for using MSC transplantation to treat cerebral infarct in the future.Result : (1) EA can improve the neural defective symptoms of MCAO rats ( P<0.05 ) . (2) EA can increase the synthesis and release of BDNF from the MCAO rats' ischemic cortx ( P<0.05 ) . (3) EA can improve the expression of NGF and Nestin in the MCAO rats' ischemic cortex (P<0.05) . (4) EA serum and the infarcted serum can't influence MSC differentiate into neuron-like cells. And DanShen injection can influence MSC differentiate into neuron-likecells.Conclusion : EA can improve the repair of the neuron damage afterthe ischemia, and EA can help to resist the damage of the brainischemia. This function maybe works by the following approach :EA can increase the synthesis and release of BDNF after theischemia , and EA can improve the expression of NGF and Nestinafter the ischmia.
