Objective: To investigate the chemical constituents of Pholidota rupestris and Eria graminifolia.Methods:(1)The chemical constituents from the EtOAc soluble fraction of P.rupestris were studied and isolated by the method of silica gel column chromatography.The structures of compounds were elucidated by spectral analyses and chemical properties.(2)Lusianthridin(Y1),coelonin(Y2),batatasin III(Y3)and thunalbene(Y4),obtained from the ethyl acetate fraction of P.rupestris,were used as standards for HPLC determination.Method validation was performed with linearity precision,stability,repeatability,selectivity,robustnes and recovery.And method validation was used to evaluate the feasibility of the method.(3)The chemical constituents from the petroleum soluble fraction of E.graminifolia were studied and isolated by the method of silica gel column chromatography.The structures of compounds were elucidated by spectral analyses and chemical properties.Results:(1)Twenty-four compounds were isolated from the ethyl acetate of 80% ethanol extract of P.rupestris and identified as confusarin(1),coelonin(2),lusianthridin(3),batatasin III(4),gigantol(5),thunalbene(6),cyclopholidone(7),cyclopholidonol(8),syringaresinol(9),?-sitosterol(10),stigamast-5-ene-3?,7?-diol(11),3,5-dimethoxy-4-hydroxypropiophenone(12),vanillin(13),4-hydroxy-3,5-dime-thoxybenzaldehyde(14),4-hydroxyphenylpropionic methyl ester(15),ochrone A(16),2,5-dimethoxybenzoquinone(17),methyl hexadecanoate(18),methyl oleate(19),decane(20),5?,8?-epidioxy-(22E,24R)-ergosta-6,22-dien-3?-ol(21),1-dodeca-nol(22),?-sitostenone(23)and ephemeranthoquinone(24).(2)The experimental results by method validation showed good linearity;the RSD% of precision was under 1.47%;the RSD% of stability was under 2.14%;the RSD% of repeatability was under 1.85%;method validation showed good selectivity and robustness;the average recoveries were in 98.98%~99.86%.And the RSD% of average recoveries was under 1.89%.The method validation results show that this method can be used to simultaneously determine the content of four compounds in the ethyl acetate of P.rupestris.(3)Seven compounds were isolated from the petroleum ether site of 75% ethanol extract of E.graminifolia and identified as decane(H1),3?-hydroxyglutin-5-ene(H2),25-hydroperoxycycloart-23-en-3?-ol(H3),erianal(H4),vanillin(H5),?-sitosterol(H6)and 2?,3?-dihydroxy propylpentadecanoate(H7).Conclusion:(1)The chemical constituents of P.rupestris were studied for the first time.Twenty-four compounds were isolated(1~24)were isolated from the EtOAc soluble fraction of P.rupestris for the first time.Among them,eight compounds(1,15~20,22)have not been recorded in the genus Pholidota previously.(2)The method simultaneously determinating of four compounds(Y1~Y4)from ethyl acetate in P.rupestris was established by HPLC-DAD for the first time.We found that four compounds mainly exist in ethyl acetate fraction of P.rupestris.The contents of four compounds in different habitats were different,but the content of batatanin III(Y3)was higher than the other three compounds.This method is accurate,simple and repeatable.And it can be used to providing a reference for the quality control of P.rupestris.(3)The chemical constituents of E.graminifolia were studied for the first time.Seven compounds(H1~H7)were isolated from the petroleum soluble fraction of E.graminifolia for the first time.Among them,four compounds(H1~H3,H7)were reporteded in the genus Eria for the first time.
