| Purpose:The changes of the number of coughs at fifth days and tenth days after the treatment of PIC rats and the changes of the content of NEP and SP in BALF and serum before and after treatment were measured by different doses of baicalen Qingfei concentrated pill.Objective to investigate the dose effect relationship of Qin Bai Qing Fei concentrated pill on improving cough symptoms and reducing immune inflammation in patients with cough after infection under NEP-SP regulatory channel.To provide the basis for the clinical use of baicalen Qingfei concentration pill in the treatment of post infection cough.Methods:1.PIC animal model was established by smoking,capsaicin induced cough combined with small dose lipopolysaccharide(LPS)to induce the infection of Gram-negative bacteria.2.The SD rats were randomly divided into 8 groups:blank group,model group,compound methoxyphenamine positive control group,low dose 1 group,low dose 2 group,middle dose 1 group,middle dose 2 group,high-dose group rats to observe the cough frequency detection of bronchoalveolar lavage NEP,and the content of SP in serum and lavage fluid(ELISA Method).Results:1.The frequency of cough: Before treatment(after modeling),except for the blank group,the cough frequency of each group was higher than 30 times.Compared with the blank group,the number of coughs in each treatment group increased significantly(P<0.05).On the fifth day after treatment,the number of coughs in each treatment group decreased,but compared with the model group,there was no significant difference between the control group,the 1 groups of baicalen low dose and the 2 groups of baicalen low dose(P>0.05),and the difference between the 1 groups,the 2 groups of baicalen middle dose and the high dose baicalen group had the significance(P<0.05).Compared with the control group,there was no significant difference in coughing times between the 1 groups of baicalen low dose,baicalen low dose 2 group and baicalen middle dose 1 group(P>0.05),and the difference of coughing times in 2 groups of baicalen middle dose and baicalen high dose group was statistically significant(P<0.05).Compared with each group,the difference between the 1 groups of baicalen low dose and 1 groups in the middle dose of baicalen,the low dose 2 groups of baicalen and the middle dose 2 groups of baicalen was statistically significant(P<0.05).There was no significant difference in the frequency of cough between the 2 groups of Qin Bai middle dose and the high dose group of Qin Bai(P>0.05).On the tenth day after treatment,Compared with the control group,the number of coughs in the high dose group was less than that of the control group(P<0.05).2.The results of SP content in the alveolar lavage fluid and serum were as follows:In BALF,the content of SP in the blank group was significantly lower than that in the model group,and the SP content in each treatment group was lower than that in the model group,and the difference was statistically significant(P<0.05).Compared with the control group,the SP content of each group was lower than that of the control group,and the difference was statistically significant(P<0.05).However,there was no significant difference between the 1 groups of baicalen middle dose and the 2 groups of baicalen middle dose(P>0.05),but the difference was statistically significant(P<0.05).There was a significant difference between the 2 groups of Qin Bai middle dose and the high dose of Qin Bai(P<0.05).In peripheral blood,the content of SP in blank group was significantly lower than that in model group.The SP content of each treatment group was lower than that of model group,and the difference was statistically significant(P<0.05).Compared with the control group,the SP content in the baicalin dose group decreased,and the difference was statistically significant(P<0.05).3.The results of NEP in the alveolar lavage fluid and serum were as follows:In BALF,the content of NEP in blank group was significantly higher than that in model group,and the difference was statistically significant(P<0.05).There was no significant difference in NEP content between control group and blank group and model group(P>0.05).There was no significant difference in the content of NEP between the 1 groups of Qin Bai low dose,compared with the model group and the control group(P>0.05).The content of NEP in the 4 groups of baicalen low dose 1 group,1 group of baicalen medium dose,baicalen medium dose 2 group and baicalen high dose group were higher than that of the model group and the control group,the difference was statistically significant(P<0.05).In peripheral blood,compared with blank group,serum NEP levels in model group,control group and Qin Bai group increased,and the difference was statistically significant(P<0.05).Compared with the model group,the content of NEP in the control group was reduced,the difference was statistically significant(P<0.05).There was no significant difference in the content of NEP in each group,and there was no statistical difference(P>0.05).Compared with the control group,the content of NEP varied in different groups,and the difference was not statistically significant(P>0.05).Conclusion:The Qin Bai Qing Fei concentrated pill can reduce the mechanism of airway neurogenic inflammation by intervening the NEP-SP pathway,effectively treat PIC cough symptoms and shorten the onset time of cough.There is a correlation between the effect and the dosage.The specific summary is as follows:1.The dosage of 1 group of Qin Bai low dose can reduce the content of SP and alleviate the inflammatory reaction outside the lesion,but this effect does not appear in the airway.There was no intervention on NEP content.So the effect of its cough is not good.2.The dosage of 2 group of Qin Bai low dose began to show the effect of increasing airway resistance and disease relief,relieving cough symptoms and preventing cough recurrence.It can be used for mild cough,but combined with other antitussive prescriptions can achieve satisfactory results.3.The dosage of 1 group of Qin Bai middle dose can reduce the airway neurogenic inflammation of PIC completely,and surpass the cough relieving effect of the control group,and can improve the ability of airway itself to resist disease and disease.4.In addition to the above effects,thedosage of 2 group of Qin Bai middle dose has the effect of shortening the onset time of cough.Its antitussive effect is the best.5.The effect of Qin Bai high dose group was slightly stronger,but it was equivalent to 2 doses of Scutellaria medium dose. |
