| Objective: Observe the changes of Wnt/β-catenin signaling pathway during the process of transdifferentiation of Shendibushen capsule in UUO rat renal tubular epithelial cells,and explore its protective mechanism of renal function and anti-renal interstitial fibrosisMethods: Sixty healthy male Sprague-Dawley rats were randomly divided into 5groups,12 in each group.They were blank group,sham operation group,model group,beinapuli group,and Shendi Bushen group.Unilateral ligation of the ureter-induced renal interstitial fibrosis model(UUO model)was used.The blank group,sham operation group,and model group were given intragastric administration of 2 ml/d distilled water,beinapuli group was given 10 mg/kg/d suspension by gavage,and Shendi Bushen group was prepared by 0.324 g/kg/d.Suspension gavage,continuous gavage for 4 weeks.Urine samples were collected at the 2nd and 4th weeks of model preparation.Serum and kidney tissue samples were collected from the anesthetized rats at the 4th week of model preparation.Observe the general state of rats;measure serum creatinine and urea nitrogen levels by enzyme method;observe urinary microalbumin changes by nephelometry;observe histological changes of kidney by HE staining;detect kidney tissue Wnt1,by immunohistochemical method Expression of β-catenin and p-GSK-3β.Decoction.:The rats in Shendi Bushen group were generally in a better condition than those in the model group;Shendi Bushen group and beinapuli group had a decrease in renal interstitial fibrosis;Shendi Bushen group and beinaepuli group had lower serum creatinine and urea nitrogen,and The model group was statistically significant(P<0.05).There was no statistically significant difference in serum creatinine and urea nitrogen between the Shenxiong kidney group and the beinapulil group(P>0.05);Shendi Bushen kidney group and beinapulil rat The decrease of microalbumin in hematuria was statistically significant compared with the model group at the same time(P<0.05).The urine microalbumin was lower in the Shendi Bushen group than in the beinapulil group(P<0.05);The expression of Wnt1,β-catenin and p-GSK-3βwere decreased in the group and beinapulil rats,which was statistically significant compared with the model group(P<0.05).The Shendi Bushen group was lower than the benazepril group.Statistical significance(P<0.05).Conclusion:1.Shendi Bushen capsule can improve the general state of rats with renal interstitial fibrosis.2.Shendi Bushen capsule can reduce the serum creatinine and urea nitrogen levels in rats with renal interstitial fibrosis,reduce the excretion of proteinuria,and delay the progression of renal failure.3.Shendi Bushen Capsule may inhibit the expression of Wnt1,p-GSK-3β and β-catenin and inhibit it the activation of Wnt/β-catenin signaling pathway delays progression of renal interstitial fibrosis. |
