Objective:To explorer the correlation between the polymorphism of MALAT1 gene rs3200401 and Type 2 diabetes mellitus(T2DM),and the correlation between basic characteristics,anthropometry and biochemical indicators between T2 DM.Methods:our research were based on the REACTION STUDY in guangxi nanning,citizens over 40 years old(living in naning more than 5 years)were selected by cluster sampling.All subjects were interviewed in questionnaire interview(including basic information,previous history,smoking history,drinking history,lifestyle and exercises),physical examination(including height,weight,waist circumference,hip circumference and blood pressure),biochemical indexes(including TC,TG,Cr,AST,ALT,FPG,2hPG,HbA1 c and FINS).Diabetes diagnosis in accordance with the diagnostic criteria of WHO1999,we used 1:1 case-control study method selected 490 T2 DM patients and 490non-diabetes controls had the same gender,age,ethnic status,smoking history,drinking history and exercises by propensity score matching.Extracted genomic DNA and detected the SNPs of MALAT1 gene rs3200401 by method of Sequenom MassArray system.Using SPSS22.0 statistical software to perform statistical analysis of correlation between the polymorphism MALAT1 gene rs3200401 and T2 DM,and analysis the were interviewed in questionnaire interview,physical examination,biochemical indexes of the studying object.Results:1.The comparison between basic information,physical examination and biochemical indexes of the two groups showed that fatty liver medical history,hypertension,history of coronary heart disease,family history of diabetes,education level,SBP,DBP,HR,Wt,WC,HC,WHR,BMI,HDL,LDL-C-C,TG,TC,FPG,2 HPG,HbA1 c,HOMA-IR and FINS had significant differences(P < 0.05).In the drinking tea,height and LDL-C has no difference between the two groups(P > 0.05).2.Univariate conditional logistic regression were used to preliminary selected the correlative factors of T2 DM,and further analysis with multinomial logistic regression.The result shows that hypertension,family history of diabetes,HR,WC and TG level were associated with increased risks of T2DM(P<0.05),and the ORs were 2.329(1.661 ~ 3.264),2.062(1.380~3.080),1.030(1.018~1.041),1.047(1.029~1.065),1.369(1.189~1.578),respectively.3.The genotypes frequencies distribution of rs3200401 between T2 DM and control groups were accorded with Hardy-Weinberg equilibrium law.The rs3200401 genotype frequencies of CC,CT and TT in the control group were78.0%,20.3% and 1.6%,in the case group were 71.9%,23.0% and 2.5%.Thefrequencies of C and T allele were 88.2% and 11.8% in the control group,and84.7% and 15.3% in the T2 DM group.We had not found any statistical differences in the genotype frequencies between T2 DM and control groups(P >0.05),but the allele frequencies between T2 DM and control groups had sinigficant differences(P < 0.05).The genotypes frequencies in recessive model of the case group were 97.5% and 2.5%,in the control group were 98.4%and 1.6%.The genotypes frequencies in dominance model of the T2 DM group were 73.8% and 26.2%,in the control group were 68.0% and 22.0%.There was no differences in recessive model between case and control groups(P > 0.05),but we found statistical differences in dominance model(P < 0.05).After adjusted for family history of diabetes,history of hypertension and triglyceride,dominance model had statistical differences.4.When compared the case with control group between different genotypes and ORs calculated with multinomial logistic regression analysis,we found no significant association between CT+TT and CC genotypes(P > 0.05).In recessive model,there was no significant association between TT and CC+CT genotypes(P > 0.05).But in dominance model,persons with CT+TT genotype carriers were associated with increased risks of T2 DM,and the ORs were 1.388([1.036~1.859],P=0.028).When compared with C allele carriers,persons with T allele carriers were associated with increased risks of T2 DM,and the ORs were 1.342([(1.034 ~ 1.741],P=0.026).Conclusion:1.Hypertension,family history of diabetes,WC and TG level were risk factors of T2 DM for Guangxi people.2.MALAT1 gene rs3200401 had correlation with T2DM: Compared with C allele and CC genotype respectively,T allele and CT+TT genotype of rs3200401 were associated with increased risks of T2 DM.
