Study On The Expression And Correlation Of Microrna-518b And Leptin In Chorionic Villi Of Missed Abortion

Objective:The main objective of the study is to investigate and analyze the difference in expression of miR-518 b and Leptin in chorionic villus of the normal pregnancy and missed abortion.Methods:1.RT-PCR technique was used to detect the expression of mi R-518 b in the chorionic villus of 13 cases of missed abortion patients(missed abortion group)and 13 cases of normal pregnant women(control group).2.The SP immunohistochemical staining method was used to detect the changes expression levels of Leptin protein in the two groups.Result:1.The RT-PCR showed the expression of miR-518 in the chorionic villus of both groups,where the expression was high in the missed abortion group than the control group and statistically significant(1.81 and 0.97,p<0.01).2.The result of SP immunohistochemical staining showed the expression of Leptin protein in the chorionic villus of two groups,mainly expressed in trophoblastic cells cytoplasm.The expression of Leptin protein in missed abortion group was significantly lower than that in the control group(P<0.01).3.Negative correlation was found between the expression of mi R-518 b and Leptin protein in the chorionic villus in both groups,i.e.the missed abortiongroup(r=-0.861,(P<0.01)),the control group(r=-0.899(P<0.01)).Conclusion:1.mi R-518 b was up-regulated in the chorionic villus of missed abortion group.2.The expression of Leptin protein in the chorionic villus of the missed abortion group is low.3.High expression of mi R-518 b in chorionic villus may be involved in the pathogenesis of missed abortion by inhibiting leptin protein synthesis.
