Objective: To study the hypotensive effects on rats and its related mechanisms induced by Bidens bipinnata L water extract.Methods:(1)Different extraction parts of permission was given to rats by the method of duodenal administration,connecting MPA multimedia biological signal recording system to measure systolic blood pressure(SBP),diastolic blood pressure(DBP)and mean arterial blood pressure(MBP)before and after the administration of medicine through carotid artery intubation.(2)After four weeks of continuous intragastric administration,the tail artery blood pressure of rats which induced by L-NAME was measured weekly.(3)The rats were killed and the blood was took when the irrigation was finished.To separate the serum and plasma of rats,the serum concerntration of nitric oxide(NO)was tested by nitric acid deoxidized enzyme.The serum concentration of T-SOD was tested by TBA method and the serumconcentration of MDA was measured by colorimetric method,the serum concentration of T-AOC was measured by hydroxylamine method,the serum concentration of norepinephrine(NE)was measured by ELISA method,the levels of endothelin 1(ET-1)and the renin-angiotensin?(Ang?)in the serum were detected by ELISA method.(4)The heart was weighed and divided into two parts.One of them was soak into 10% formaldehyde solution for routine pathological sections,HE staining and immunohistochemistry(SP method)in order to determine the expression of NF-?B in myocardium,TNF-?,PPAR-?,collagen ?,collagen ? protein expression differences.The expression of NF-?B and TNF-? mRNA in the myocardium of each group was detected by read-PCR.Results:(1)Bidens bipinnata L water extract makes rats and L-NAME induced hypertensive rats decreased significantly(P <0.01).(2)After four weeks of irrigation for hypertensive rats induced by L-NAME from all the treatment groups which can be observed that: one week after the administration,the SBP and DBP of captopril group began to decline,after the last administration,the SBP and DBP of captopril group,persimmon root water extract high-dose group,middle dose group and the low dose group were significantly decreased(P <0.01).The rats had steady increases in body weight,with no apparent difference between the groups(P>0.05).(3)Compared of the model group,the content of NO,the activities of TSOD and T-AOC were increased significantly(P <0.05 or P <0.01)in each treatment group,meanwhile the content of MDA,NE in the serum,the contentof ET-1,Ang ? in the plasma were decreased significantly(P <0.05 or P<0.01).(4)The heart HE staining showed that the tissue of model group disorder,break,dissolve,cardiomyocyte hypertrophy,and each treatment group of persimmon root aqueous extract was significantly reduced compared with the model group.(5)Connective tissue growth factor NF-?B,TNF-?,PPAR-?,collagen ?,collagen ? protein relative expression results;the immunohistochemical results showed cytoplasm and nucleus was dyed yellow and brown as the positive expression of NF-?B,TNF-?,PPAR-?,collagen ?,collagen ?.Compared with blank control group,model control group the positive expression range increased.High dose Bidens bipinnata L groups were compared with the model control group of the positive expression range decrease(P<0.01).(6)Connective tissue growth factor NF-?B,TNF-? gene relative expression results: compared with blank control group,model control group NF-?B,TNF-?gene expression quantity increase(P<0.01).Compared with model control group,high dose Bidens bipinnata L group of NF-?B,TNF-? gene expression quantity reduced(P<0.01).Conclusions:(1)Water extract of Bidens bipinnata L can reduce blood pressure in normal rats and L-NAME induced hypertension in rats.(2)Water extract of Bidens bipinnata L significantly relieve left ventricular hypertrophy in hypertensive rats with myocardial fibrosis effect,and its antihypertention mechanism may be due to increase the production and releaseof NO,to reduce the production of the endogenous vasoconstrictor and vascular active substances which called NE,Ang? and ET-1.(3)Water extract of Bidens bipinnata L can significantly relieve the disorder,dissolution,pathological fracture,fibrillation and other injuries of the cardiac.(4)Bidens bipinnata L groups can reduce NF-?B,TNF-?,PPAR-?,collagen ?,collagen ? gene expression of protein(5)Bidens bipinnata L groups can reduce NF-?B,TNF-? gene expression of mRNA.
